Western Blot (WB) (Detection of Mouse and Rat Brorin/VWC2 by Western Blot. Western blot shows lysates of rat cortical neuron tissue and mouse brain (thalamus/hypothalamus) tissue. PVDF Membrane was probed with 1ug/ml of Sheep Mouse Brorin/VWC2 Antigen Affinity­ purified Polyclonal Antibody followed by HRP­ conjugated Sheep IgG Secondary Antibody. A specific band was detected for Brorin/VWC2 at approximately 35kD (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.)

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Immunohistochemistry (IHC)

Brorin/VWC2 was detected in immersion fixed frozen sections of mouse embryo (13 d.p.c.) using Sheep Mouse Brorin/VWC2 Antigen Affinity­purified Polyclonal Antibody at 5ug/ml overnight at 4 °C. Tissue was stained using the Sheep HRP­DAB Cell & Tissue Staining Kit a n d counterstained with hematoxylin (blue). Specific staining was localized to the cytoplasm of mesenchyme/muscle cells. Mouse VWC2 is a secreted, 35kD (predicted) member of the chordin family of proteins. It is expressed in the choroid plexus and by neurons in multiple sites, and serves to sequester BMP family proteins such as BMP-2 and BMP-6. BMP antagonism is suggested to generate neurons from neural precursor cells at the expense of astrocytes. Mouse VWC2 precursor is 324aa in length. It contains a 27aa signal sequence, one poly-Ala region (aa124-129), and two von Willebrand factor C domains (aa152-211, 215-273) that likely serve as BMP interaction sites. Over aa28-324, mouse VWC2 shares 90aa identity with human VWC2.

Uniprot Description

Function: BMP antagonist which may play a role in neural development. Promotes cell adhesion. Ref.1 Ref.7

Subunit structure: Peripherally associated with AMPAR complex. AMPAR complex consists of an inner core made of 4 pore-forming GluA/GRIA proteins (GRIA1, GRIA2, GRIA3 and GRIA4) and 4 major auxiliary subunits arranged in a twofold symmetry. One of the two pairs of distinct binding sites is occupied either by CNIH2, CNIH3 or CACNG2, CACNG3. The other harbors CACNG2, CACNG3, CACNG4, CACNG8 or GSG1L. This inner core of AMPAR complex is complemented by outer core constituents binding directly to the GluA/GRIA proteins at sites distinct from the interaction sites of the inner core constituents. Outer core constituents include at least PRRT1, PRRT2, CKAMP44/SHISA9, FRRS1L and NRN1. The proteins of the inner and outer core serve as a platform for other, more peripherally associated AMPAR constituents, including VWC2. Alone or in combination, these auxiliary subunits control the gating and pharmacology of the AMPAR complex and profoundly impact their biogenesis and protein processing. Ref.8

Tissue specificity: Predominantly expressed in the brain (at protein level). It is expressed in the neurons but not the glial cells. Ref.1 Ref.7 Ref.8

Developmental stage: At 12.5 dpc, predominantly expressed in the developing diencephalon. At 16.5 dpc and 18.5 dpc, expressed in the brain, spinal cord, developing neural tubes and tongue but not in the cerebral cortex. At 16.5 dpc, present in developing oral and tooth germ epithelia (at protein level). Ref.1 Ref.7
