Vang-like protein 1 (VANGL1), also called Loop-tail protein 2 homolog (LPP2), is encoded by the VANGL1 gene, This gene contains 8 exons[1] and it maps to chromosome 1p13[2]. The VANGL1 protein is a 524-amino acid, 65KD protein, which has 4 transmembrane domains and a C-terminal ser/thr-x-val motif. Yagyu et al. (2002) found that downregulation of VANGL1 expression suppressed growth of a human hepatoma cell line.[1] Kalabis et al. (2006) found that VANGL1 overexpression enhanced this migration and wound closure in cultured cells.[3]

This gene encodes a member of the tretraspanin family. The encoded protein may be involved in mediating intestinal trefoil factor induced wound healing in the intestinal mucosa. Mutations in this gene are associated with neural tube defects. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Feb 2010]

Involvement in disease: Neural tube defects (NTD) [MIM:182940]: Congenital malformations of the central nervous system and adjacent structures related to defective neural tube closure during the first trimester of pregnancy. Failure of neural tube closure can occur at any level of the embryonic axis. Common NTD forms include anencephaly, myelomeningocele and spina bifida, which result from the failure of fusion in the cranial and spinal region of the neural tube. NTDs have a multifactorial etiology encompassing both genetic and environmental components.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.11 Ref.12Sacral defect with anterior meningocele (SDAM) [MIM:600145]: Form of caudal dysgenesis. It is present at birth and becomes symptomatic later in life, usually because of obstructive labor in females, chronic constipation, or meningitis.
