UVRAG is predicted to contain a coiled-coil structure and is likely to be metabolically unstable based on the occurrence of a strong PEST domain. It interacts with Beclin 1, leading to activation of autophagy and thereof inhibition of tumorigenicity of human colon cancer cells. The gene for UVRAG protein is localized on human chromosome 11q13.5 and complements theultraviolet sensitivity of xeroderma pigmentosum group C cells. The gene has been extremely well conserved during evolution, implying an important role for this gene product in cell metabolism. Chromosomal aberrations involving this gene are associated with left-right axis malformation, and mutations in this gene have been associated with colon cancer. The protein is highly expressed in human brain, lung, kidney and liver.

NCBI Description

This gene complements the ultraviolet sensitivity of xeroderma pigmentosum group C cells and encodes a protein with a C2 domain. The protein activates the Beclin1-PI(3)KC3 complex, promoting autophagy and suppressing the proliferation and tumorigenicity of human colon cancer cells. Chromosomal aberrations involving this gene are associated with left-right axis malformation and mutations in this gene have been associated with colon cancer. [provided by RefSeq, Jul 2008]

Subunit structure: Forms a complex with BECN1, PIK3C3 and PIK3R4. A subpopulation of these complexes also harbor KIAA0226/Rubicon. UVRAG-containing complexes never including ATG14, both forming mutually exclusive complexes with BECN1 through direct competition. Directly interacts with FEZ1 and SCOC. The interaction with SCOC is reduced by amino acid starvation, but the complex is stabilized in the presence of FEZ1. Interacts with BECN1P1/BECN2. Ref.3 Ref.6 Ref.8 Ref.11 Ref.12

Subcellular location: Late endosome. Lysosome. Early endosome. Note: Colocalizes with RAB9-positive compartments involved in retrograde transport from late endosomes to trans-Golgi network. Colocalization with early endosomes is only partial. Ref.3

Tissue specificity: Highly expressed in brain, lung, kidney and liver. Ref.2

Involvement in disease: A chromosomal aberration involving UVRAG has been observed in a patient with heterotaxy (left-right axis malformation). Inversion Inv11(q13.5;q25).

Sequence similarities: Contains 1 C2 domain.
