Dilution: ELISA: 1:1,000
Western Blot: 1:100-500
Immunohistochemistry: 1:50-100
Flow Cytometry: 1:10-50
Western Blot (WB)
(Western blot analysis of TNPO1 Antibody (N-term) in 293 cell line lysates (35ug/lane). TNPO1 (arrow) was detected using the purified Pab.)
Immunohistochemistry (IHC)
(Formalin-fixed and paraffin-embedded human brain tissue reacted with TNPO1 Antibody (N-term), which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated.)
Flow Cytometry (FC/FACS)
(TNPO1 Antibody (N-term) flow cytometry analysis of 293 cells (bottom histogram) compared to a negative control cell (top histogram).FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.)
NCBI and Uniprot Product Information
Uniprot Description
Function: Functions in nuclear protein import as nuclear transport receptor. Serves as receptor for nuclear localization signals (NLS) in cargo substrates. Is thought to mediate docking of the importin/substrate complex to the nuclear pore complex (NPC) through binding to nucleoporin and the complex is subsequently translocated through the pore by an energy requiring, Ran-dependent mechanism. At the nucleoplasmic side of the NPC, Ran binds to the importin, the importin/substrate complex dissociates and importin is re-exported from the nucleus to the cytoplasm where GTP hydrolysis releases Ran. The directionality of nuclear import is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus
By similarity. Involved in nuclear import of M9-containing proteins. In vitro, binds directly to the M9 region of the heterogeneous nuclear ribonucleoproteins (hnRNP), A1 and A2 and mediates their nuclear import. Appears also to be involved in hnRNP A1/A2 nuclear export. Mediates the nuclear import of ribosomal proteins RPL23A, RPS7 and RPL5. Binds to a beta-like import receptor binding (BIB) domain of RPL23A
By similarity. In vitro, mediates nuclear import of SRP19
By similarity and H2A, H2B, H3 and H4 histones. Mediates nuclear import of ADAR/ADAR1 in a RanGTP-dependent manner
By similarity. Ref.3
Subunit structure: Interacts with H2A, H2B, H3 and H4 histones
By similarity. Binds HNRPA1, HNRPA2, HNRNPDL, RPL23A, RPS7, RPL5, RAN and SRP19. Interacts with ADAR/ADAR1 (via DRBM 3 domain). Interacts with SNAI1 (via zinc fingers); the interaction mediates SNAI1 nuclear import. Interacts with SNAI2 (via zinc fingers)
By similarity. Ref.3
Subcellular location: Cytoplasm
By similarity. Nucleus
By similarity.
Sequence similarities: Belongs to the importin beta family.Contains 8 HEAT repeats.Contains 1 importin N-terminal domain.
Sequence caution: The sequence BAC26696.1 differs from that shown. Reason: Erroneous initiation. The sequence BAC27029.1 differs from that shown. Reason: Erroneous initiation.

Product Notes
The Tnpo1 tnpo1 (Catalog #AAA648790) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The TNPO1, NT (TNPO1, KPNB2, MIP1, TRN, Transportin-1, Importin beta-2, Karyopherin beta-2, M9 region interaction protein) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's TNPO1 can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Immunohistochemistry (IHC), Flow Cytometry (FC/FACS). Suitable for use in Western Blot, Immunohistochemistry, Flow Cytometry, ELISA Dilution: ELISA: 1:1,000 Western Blot: 1:100-500 Immunohistochemistry: 1:50-100 Flow Cytometry: 1:10-50. Researchers should empirically determine the suitability of the Tnpo1 tnpo1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TNPO1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
