Related Product Information for anti-TLR4 antibody
Background: TLR4 (Toll-like receptor 4) is a mammalain homologue of the Drosophila toll protein. The human TLR4 is a type-1 transmembrane protein containing 799 amino acid residues. The extracellular domain has leucine-rich repeats and the intracellular domain is similar to that of IL-1 receptor. TLR4 is a receptor for lipopolysaccharide (LPS). In mice that are genetically tolerant to endotoxin, mutations have been identified in the intracellular domain of TLR4 that abolish LPS responsiveness. TLR4 uses a mechanism similar to that of the IL-1 receptor for signal transduction that leads to activation of NF-kB.

NCBI Description

The protein encoded by this gene is a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This receptor has been implicated in signal transduction events induced by lipopolysaccharide (LPS) found in most gram-negative bacteria. Mutations in this gene have been associated with differences in LPS responsiveness. Multiple transcript variants encoding different isoforms have been found for this gene. Uniprot Description

TLR4: Cooperates with LY96 and CD14 to mediate the innate immune response to bacterial lipopolysaccharide (LPS). Acts via MYD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Also involved in LPS- independent inflammatory responses triggered by Ni(2+). These responses require non-conserved histidines and are, therefore, species-specific. Belongs to the lipopolysaccharide (LPS) receptor, a multi-protein complex containing at least CD14, LY96 and TLR4. Binding to bacterial LPS leads to homodimerization. Interacts with LY96 via the extracellular domain. Interacts with MYD88 and TIRAP via their respective TIR domains. Interacts with NOX4. Interacts with CNPY3. Interacts with HSP90B1. The interaction with both CNPY3 and HSP90B1 is required for proper folding in the endoplasmic reticulum. Highly expressed in placenta, spleen and peripheral blood leukocytes. Detected in monocytes, macrophages, dendritic cells and several types of T-cells. Belongs to the Toll-like receptor family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The TLR4 tlr4 (Catalog #AAA840411) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The TLR4 Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's TLR4 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Flow Cytometry (FC/FACS). Western blot analysis (0.5-4 ug/ml). The antibody is also suitable for flow cytometric analysis of cell surface expression of TLR4. However, the optimal conditions should be determined individually. Researchers should empirically determine the suitability of the TLR4 tlr4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TLR4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
