NCBI Description

This gene encodes a protease inhibitor that regulates the tissue factor (TF)-dependent pathway of blood coagulation. The coagulation process initiates with the formation of a factor VIIa-TF complex, which proteolytically activates additional proteases (factors IX and X) and ultimately leads to the formation of a fibrin clot. The product of this gene inhibits the activated factor X and VIIa-TF proteases in an autoregulatory loop. The encoded protein is glycosylated and predominantly found in the vascular endothelium and plasma in both free forms and complexed with plasma lipoproteins. Several alternatively spliced transcript variants of this gene have been described, but the full-length nature of some of these variants has not been confirmed. [provided by RefSeq]

Uniprot Description

Function: Inhibits factor X (X(a)) directly and, in a Xa-dependent way, inhibits VIIa/tissue factor activity, presumably by forming a quaternary Xa/LACI/VIIa/TF complex. It possesses an antithrombotic action and also the ability to associate with lipoproteins in plasma.

Subcellular location: Secreted.

Tissue specificity: Mostly in endothelial cells.

Domain: This inhibitor contains three inhibitory domains. The first domain interacts with VIIa and TF, the second one with Xa.

Post-translational modification: O-glycosylated. Ref.10 Ref.14 Ref.16

Sequence similarities: Contains 3 BPTI/Kunitz inhibitor domains.

Product Notes

The TFPI tfpi (Catalog #AAA512162) is an Antibody produced from Sheep and is intended for research purposes only. The product is available for immediate purchase. This product is suitable as a source of antibodies to TFPI. Researchers should empirically determine the suitability of the TFPI tfpi for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Tissue Factor Pathway Inhibitor, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
