Western Blot (WB) (Figure: Western Blot detection of endogenous and overexpressed TIGAR using PAb to TIGAR (AT122) . Method: Cell extracts from HEK 293 (lane 1), HEK 293T (lane 2) or HEK 293T transfected with a plasmid coding for human TIGAR (lane 3) were resolved by SDS-PAGE under reducing conditions, transferred to nitrocellulose and incubated with the PAb to TIGAR (AT122) at a 1:1'000 dilution for 1 hour. Proteins were visualized using a peroxidase-conjugated PAb to rabbit IgG and a chemiluminescence detection system.)

Related Product Information for anti-TIGAR antibody
The p53 tumor-suppressor protein prevents cancer development through various mechanisms, including the induction of cell-cycle arrest, apoptosis, and the maintenance of genome stability. TIGAR (TP53-induced glycolysis and apoptosis regulator) has been identified as a p53-inducible gene. TIGAR expression decreased fructose-2,6-bisphosphate levels in cells, resulting in an inhibition of glycolysis. In addition, TIGAR expression lowered intracellular reactive oxygen species (ROS) levels and protected cells from p53-induced apoptosis. Bensaad et al., proposed that TIGAR modulates the apoptotic response to p53, thereby allowing cells to survive mild or transient stress.

NCBI Description

This gene is regulated as part of the p53 tumor suppressor pathway and encodes a protein with sequence similarity to the bisphosphate domain of the glycolytic enzyme that degrades fructose-2,6-bisphosphate. The protein functions by blocking glycolysis and directing the pathway into the pentose phosphate shunt. Expression of this protein also protects cells from DNA damaging reactive oxygen species and provides some protection from DNA damage-induced apoptosis. The 12p13.32 region that includes this gene is paralogous to the 11q13.3 region. [provided by RefSeq, Jul 2008]

Uniprot Description

TIGAR: Fructose-bisphosphatase hydrolyzing fructose-2,6- bisphosphate as well as fructose-1,6-bisphosphate. Inhibits glycolysis by reducing cellular levels of fructose-2,6- bisphosphate. May protect cells against reactive oxygen species and against apoptosis induced by tp53. Belongs to the phosphoglycerate mutase family.

Protein type: EC 3.1.3.46; Phosphatase (non-protein)

Chromosomal Location of Human Ortholog: 12p13.3

Cellular Component: cytoplasm; cytosol; intracellular; mitochondrial outer membrane; nucleus

Molecular Function: bisphosphoglycerate phosphatase activity; fructose-2,6-bisphosphate 2-phosphatase activity; protein binding

Biological Process: apoptosis; autophagy; dephosphorylation; fructose 1,6-bisphosphate metabolic process; fructose 2,6-bisphosphate metabolic process; gene expression; negative regulation of glycolysis; negative regulation of programmed cell death; positive regulation of DNA repair; regulation of pentose-phosphate shunt; response to DNA damage stimulus; response to gamma radiation; response to xenobiotic stimulus; transcription initiation from RNA polymerase II promoter

Product Notes

The TIGAR tigar (Catalog #AAA565341) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The TIGAR polyclonal antibody (AT122) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's TIGAR can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Western Blot (1:1,000) Suggested dilutions/conditions may not be available for all applications. Does not work for IP. Researchers should empirically determine the suitability of the TIGAR tigar for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TIGAR, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
