SDS-Page (Peptide Competition Extracts prepared from HEK293 cells left untreated (1) or treated with UV (2-5) were resolved by SDS-PAGE on a 10% polyacrylamide gel and transferred to PVDF. Membranes were blocked with a 5% BSA-TBST buffer for one hour at room temperature, then incubated with STAT1 [pS727] antibody for two hours at room temperature in a 3% Milk-TBST buffer, following prior incubation with: no peptide (1, 2), the non-phosphopeptide corresponding to the immunogen (3), a generic phosphoserine containing peptide (4), or, the phosphopeptide immunogen (5). After washing, membranes were incubated with goat F(ab’)2 anti-rabbit IgG HRPconjugate and bands were detected using the Pierce SuperSignal method. The data show that only the peptide corresponding to STAT1 [pS727] blocks the antibody signal, demonstrating the specificity of the antibody.)

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

The signal transducer and activator of transcription (STAT) family of transcription factors is composed of seven family members (STATs 1, 2, 3, 4, 5A, 5B and 6) that are involved in many cell processes (apoptosis, anti-apoptosis, cell differentiation and proliferation) depending upon the cell type. STATs are activated by ligand binding of mostly cytokine receptors, that receptor associating with a Janus kinase (JAK) family member that phosphorylates a STAT protein causing it to translocate to the nucleus, where it can bind to DNA and regulate the transcription of particular genes. STAT1 serine 727 can be phosphorylated by p38, PKC and CaMKII and has been found to be required for maximum transcriptional activity.

NCBI Description

The protein encoded by this gene belongs to the acyltransferase family. It is located in the endoplasmic reticulum, and catalyzes the formation of fatty acid-cholesterol esters. This gene has been implicated in the formation of beta-amyloid and atherosclerotic plaques by controlling the equilibrium between free cholesterol and cytoplasmic cholesteryl esters. Alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Nov 2011]
