Biological Significance
SPPL2B is a member of the GXGD family of aspartic proteases. The GXGD proteases are transmembrane proteins with two conserved catalytic motifs localized within the membrane-spanning regions. This enzyme localizes to endosomes, lysosomes, and the plasma membrane. It cleaves the transmembrane domain of tumor necrosis factor alpha to release the intracellular domain, which triggers cytokine expression in the innate and adaptive immunity pathways.
Cross-Reactivity
Human
Immunogen
Synthetic peptide directed toward the N terminal region of human SPPL2B.
Preparation and Storage
Ships ambient or refrigerated. Upon receipt store at 4°C for short term, -20°C for long term.

NCBI Description

This gene encodes a member of the GXGD family of aspartic proteases. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Uniprot Description

SPPL2B: Intramembrane-cleaving aspartic protease (I-CLiP) that cleaves type II membrane signal peptides in the hydrophobic plane of the membrane. Functions in ITM2B and TNF processing. Catalyzes the intramembrane cleavage of the anchored fragment of shed TNF- alpha (TNF), which promotes the release of the intracellular domain (ICD) for signaling to the nucleus. May play a role in the regulation of innate and adaptive immunity. Belongs to the peptidase A22B family. 4 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, multi-pass; EC 3.4.23.-; Membrane protein, integral; Protease

Chromosomal Location of Human Ortholog: 19p13.3

Cellular Component: lysosomal membrane; plasma membrane; endosome membrane

Molecular Function: protein binding; protein homodimerization activity; aspartic endopeptidase activity, intramembrane cleaving

Biological Process: regulation of immune response; membrane protein intracellular domain proteolysis; membrane protein ectodomain proteolysis

Product Notes

The SPPL2B sppl2b (Catalog #AAA839114) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The SPPL2B antibody reacts with Cow, Dog, Guinea Pig, Horse, Human, Mouse, Pig, Rat. and may cross-react with other species as described in the data sheet. AAA Biotech's SPPL2B can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC). WB: 1 ug/ml IHC: 4-8 ug/ml. Researchers should empirically determine the suitability of the SPPL2B sppl2b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SPPL2B, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
