Purified by Protein A affinity chromatography.
Dilution: ELISA: 1:1,000
Western Blot: 1:100-500
Immunohistochemistry: 1:10-50
Immunofluorescence (IF)
(Fluorescent image of NIH/3T3 cell stained with SPI1 Antibody (C-term) .NIH/3T3 cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with SPI1 primary antibody (1:25, 1 h at 37℃). For secondary antibody, Alexa Fluor® 488 conjugated donkey anti-rabbit antibody (green) was used (1:400, 50 min at 37℃).Cytoplasmic actin was counterstained with Alexa Fluor® 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37℃). SPI1 immunoreactivity is localized to Nucleus significantly.)
Western Blot (WB)
(SPI1 Antibody (C-term) western blot analysis in WiDr cell line lysates (35ug/lane).This demonstrates the SPI1 antibody detected the SPI1 protein (arrow).)
Immunohistochemistry (IHC)
(SPI1 Antibody (C-term) (AP14850b)immunohistochemistry analysis in formalin fixed and paraffin embedded human tonsil tissue followed by peroxidase conjugation of the secondary antibody and DAB staining. This data demonstrates the use of SPI1 Antibody (C-term) for immunohistochemistry. Clinical relevance has not been evaluated.)
NCBI and Uniprot Product Information
NCBI Description
This gene encodes an ETS-domain transcription factor that activates gene expression during myeloid and B-lymphoid cell development. The nuclear protein binds to a purine-rich sequence known as the PU-box found near the promoters of target genes, and regulates their expression in coordination with other transcription factors and cofactors. The protein can also regulate alternative splicing of target genes. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
Uniprot Description
PU.1: Binds to the PU-box, a purine-rich DNA sequence (5'- GAGGAA-3') that can act as a lymphoid-specific enhancer. This protein is a transcriptional activator that may be specifically involved in the differentiation or activation of macrophages or B- cells. Also binds RNA and may modulate pre-mRNA splicing. Binds DNA as a monomer. Interacts with CEBPD and NONO. Interacts with RUNX1 and SPIB. Interacts with GFI1; the interaction represses SPI1 transcriptional activity. Highly expressed in both FV-P and FV-A-induced erythro- leukemia cell lines that have undergone rearrangements of the Spi- 1 gene due to the insertion of SFFV. Belongs to the ETS family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes
The SPI1 spi1 (Catalog #AAA6010804) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The SPI1, CT (SPI1, Transcription factor PU.1, 31kD-transforming protein) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's SPI1 can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Immunohistochemistry (IHC). Suitable for use in Western Blot, Immunohistochemistry, ELISA Dilution: ELISA: 1:1,000 Western Blot: 1:100-500 Immunohistochemistry: 1:10-50. Researchers should empirically determine the suitability of the SPI1 spi1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SPI1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
