NCBI Description

This gene encodes a protein with several transmembrane domains, a Rab11-binding domain and a lipid-binding FYVE finger domain. The encoded protein appears to promote neurite formation. A mutation in this gene has been reported to be associated with hereditary spastic paraplegia, however the pathogenicity of the mutation, which may simply represent a polymorphism, is unclear. [provided by RefSeq, Mar 2010]

Uniprot Description

ZFYVE27: Functions as an upstream inhibitor of RAB11, regulating directional protein transport to the forming neurites. Involved in nerve growth factor-induced neurite formation. May have a more general role in cell projections formation. Defects in ZFYVE27 are the cause of spastic paraplegia autosomal dominant type 33 (SPG33). Spastic paraplegia is a neurodegenerative disorder characterized by a slow, gradual, progressive weakness and spasticity of the lower limbs. Rate of progression and the severity of symptoms are quite variable. Initial symptoms may include difficulty with balance, weakness and stiffness in the legs, muscle spasms, and dragging the toes when walking. In some forms of the disorder, bladder symptoms (such as incontinence) may appear, or the weakness and stiffness may spread to other parts of the body. According to PubMed:18606302, the properties of the variant Val-191 and its frequency in some populations raise doubts on the implication of that gene in the disease. 5 isoforms of the human protein are produced by alternative splicing.

Product Notes

The SPG33 zfyve27 (Catalog #AAA416700) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-SPG33 Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's SPG33 can be used in a range of immunoassay formats including, but not limited to, Peptide ELISA (EIA), Western Blot (WB). Peptide ELISA titer: 1:40,000-1:80,000 Western blot analysis: 1:200-500 Optimal dilutions/concentrations should be determined by the end user. Researchers should empirically determine the suitability of the SPG33 zfyve27 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SPG33, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
