Immunofluorescence (IF) (Fluorescent confocal image of HeLa cells stained with phospho-SMAD3-S208 antibody. HeLa cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with AP3249a phospho-SMAD3-S208 primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (5.25 uM, 25 min). Pictures were taken on a Biorevo microscope (BZ-900, Keyence).Note the highly specific localization of the phospho-SMAD3 mainly to the nucleus, supported by Human Protein Atlas Data.)

NCBI Description

The protein encoded by this gene belongs to the SMAD, a family of proteins similar to the gene products of the Drosophila gene 'mothers against decapentaplegic' (Mad) and the C. elegans gene Sma. SMAD proteins are signal transducers and transcriptional modulators that mediate multiple signaling pathways. This protein functions as a transcriptional modulator activated by transforming growth factor-beta and is thought to play a role in the regulation of carcinogenesis. [provided by RefSeq, Apr 2009]

Product Notes

The SMAD3 smad3 (Catalog #AAA6499811) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The SMAD3, Phosphorylated (Ser208) (Mothers Against Decapentaplegic Homolog 3, SMAD 3, Mothers Against DPP Homolog 3, MAD Homolog 3, Mad3, hMAD-3, SMAD Family Member 3, JV15-2, hSMAD3, MADH3) (Biotin) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's SMAD3 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Dot Blot (DB), Immunofluorescence (IF). ELISA: 1:1,000 Dot Blot: 1:500 IF: 1:10-1:50 Applications are based on unconjugated antibody. Researchers should empirically determine the suitability of the SMAD3 smad3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SMAD3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
