Goat Silver homologue/Pmel 17 Polyclonal Antibody | anti-SILV antibody
Goat anti-Silver homologue/Pmel 17 (C-terminus) Antibody
Western Blot: Approx 26kDa band observed in Human Skin lysates, corresponding to the M-beta fragment of the precursor protein (Leonhardt et al, Mol Biol Cell.2013 Apr;24(7): 964-81. PMID: 23389629). Calculated MW of 70.3kDa according to NP_008859.1. In transfected HEK293 transiently expressing Human SILV a band of approx.70kDa is observed. This band is not observed in the non-transfected HEK293. Recommended concentration: 1-3ug/ml.
Western Blot (WB)
((1ug/ml) staining of Human Skin lysate (35ug protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.)
Western Blot (WB)
(HEK293 overexpressing Human SILV (RC200663) and probed (mock transfection in first lane), tested by Origene, data obtained from previous batch (different goat).)
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a melanocyte-specific type I transmembrane glycoprotein. The encoded protein is enriched in melanosomes, which are the melanin-producing organelles in melanocytes, and plays an essential role in the structural organization of premelanosomes. This protein is involved in generating internal matrix fibers that define the transition from Stage I to Stage II melanosomes. This protein undergoes a complex pattern of prosttranslational processing and modification that is essential to the proper functioning of the protein. A secreted form of this protein that is released by proteolytic ectodomain shedding may be used as a melanoma-specific serum marker. Alternate splicing results in multiple transcript variants. Uniprot Description
SILV: Plays a central role in the biogenesis of melanosomes. Involved in the maturation of melanosomes from stage I to II. The transition from stage I melanosomes to stage II melanosomes involves an elongation of the vesicle, and the appearance within of distinct fibrillar structures. Release of the soluble form, ME20-S, could protect tumor cells from antibody mediated immunity. Belongs to the PMEL/NMB family. 5 isoforms of the human protein are produced by alternative splicing.
Protein type: Membrane protein, integral; Vesicle
Chromosomal Location of Human Ortholog: 12q13-q14
Cellular Component: endoplasmic reticulum membrane; extracellular region; Golgi apparatus; integral to plasma membrane; melanosome; plasma membrane
Molecular Function: protein binding
Biological Process: melanin biosynthetic process; melanosome organization and biogenesis; pigmentation during development

The Goat anti-Silver homologue/Pmel 17 (C-terminus) Antibody reacts with Tested: Human; Expected from sequence similarity: Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Silver homologue/Pmel 17 can be used in a range of immunoassay formats including, but not limited to, Peptide ELISA (EIA), Western Blot (WB). Peptide ELISA: Antibody detection limit dilution 1: 128000. Western Blot: Approx 26kDa band observed in Human Skin lysates, corresponding to the M-beta fragment of the precursor protein (Leonhardt et al, Mol Biol Cell.2013 Apr;24(7): 964-81. PMID: 23389629). Recommended concentration: 1-3ug/ml. Researchers should empirically determine the suitability of the SILV pmel for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: CPIGENSPLL SGQQ. It is sometimes possible for the material contained within the vial of "Silver homologue/Pmel 17, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
