Western Blot (WB)
(Western blot analysis of extracts from Hela, using SIK (Phospho-Thr182) Antibody.)
Western Blot (WB)
(Western blot analysis SIK (Phospho-Thr182) using EGF treated HeLa whole cell lysates.-/+ means absence or presence of N peptide(non-phospho peptide) and P peptide(phospho peptide).)
Immunohistochemistry (IHC)
(At 1/200 staining Rat liver tissue sections by IHC-P. The tissue was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The tissue was then blocked and incubated with the antibody for 1.5 hours at 22 degree C. An HRP conjugated goat anti-rabbit antibody was used as the secondary antibody.)
Immunohistochemistry (IHC)
(At 1/200 staining Mouse testis tissue sections by IHC-P.
(At 1/200 staining Human lung cancer tissue sections by IHC-P.
NCBI and Uniprot Product Information
Uniprot Description
SIK: Serine/threonine-protein kinase involved in various processes such as cell cycle regulation, gluconeogenesis and lipogenesis regulation, muscle growth and differentiation and tumor suppression. Phosphorylates HDAC4, HDAC5, PPME1, SREBF1, TORC1/CRTC1 and TORC2/CRTC2. Acts as a tumor suppressor and plays a key role in p53/TP53-dependent anoikis, a type of apoptosis triggered by cell detachment: required for phosphorylation of p53/TP53 in response to loss of adhesion and is able to suppress metastasis. Part of a sodium-sensing signaling network, probably by mediating phosphorylation of PPME1: following increases in intracellular sodium, SIK1 is activated by CaMK1 and phosphorylates PPME1 subunit of protein phosphatase 2A (PP2A), leading to dephosphorylation of sodium/potassium-transporting ATPase ATP1A1 and subsequent increase activity of ATP1A1. Acts as a regulator of muscle cells by phosphorylating and inhibiting class II histone deacetylases HDAC4 and HDAC5, leading to promote expression of MEF2 target genes in myocytes. Also required during cardiomyogenesis by regulating the exit of cardiomyoblasts from the cell cycle via down-regulation of CDKN1C/p57Kip2. Acts as a regulator of hepatic gluconeogenesis by phosphorylating and repressing the CREB-specific coactivators TORC1/CRTC1 and TORC2/CRTC2, leading to inhibit CREB activity. Also regulates hepatic lipogenesis by phosphorylating and inhibiting SREBF1. Interacts with ATP1A1. Interacts (when phosphorylated on Thr-182 and Ser-186) with YWHAZ. Activated by phosphorylation on Thr-182. Also activated by phosphorylation on Thr-322 in response to increases in intracellular sodium in parallel with elevations in intracellular calcium through the reversible sodium/calcium exchanger. Belongs to the protein kinase superfamily. CAMK Ser/Thr protein kinase family. AMPK subfamily.

Product Notes
The SIK sik1 (Catalog #AAA9614190) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Phospho-SIK (Thr182) Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig (100%), Zebrafish (90%), Dog (100%), Chicken (100%) and may cross-react with other species as described in the data sheet. AAA Biotech's SIK can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC), Peptide ELISA (EIA). WB: 1:1000-3000 IHC: 1:50-1:200 Peptide ELISA: 1:20,000-1:40,000. Researchers should empirically determine the suitability of the SIK sik1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SIK, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
