Western Blot (WB) (Detection of Human Phospho­RPA2 (T21) by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line and U2OS human osteosarcoma cell line untreated (­ ) or treated (+) with 1 μM Camptothecin (CPT) for 1 hour. PVDF Membrane was probed with 0.5ug/ml of Human Phospho­RPA2 (T21) Antigen Affinity­ purified Polyclonal Antibody followed by HRP­conjugated Rabbit IgG Secondary Antibody. A specific band was detected for Phospho­RPA2 (T21) at approximately 40kD (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.)

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

RPA2 is a 32kD DNA-binding protein that constitutess one of three subunits comprising the PRA heterotrimer complex. In conjunction with 70kD RPA1 and 14kD RPA3, RPA2 participates in DNA replication, recombination and repair. Human RPA2 is 270aa in length. It contains a Gly/Ser-rich N-terminus (aa1-33), a DNA-binding domain (aa43-171) and a protein-interaction C-terminus (aa187-270). Phosphorylation of the N-terminus on Ser4/8/23/29/33, plus Thr21, regulates RPA complex interactions with DNA repair and replication complexes. There are multiple splice variants. Three contain N-terminal extensions: one shows an 88aa insertion after Ser4, another shows a 12aa substitution for aa1-4, and a third shows a four aa insertion after Ser4. There is also a deletion of aa93-98, and a potential truncation after Gln175. Over aa141-270, human RPA2 shares 83% aa identity with mouse RPA2.

RFA2: Required for DNA recombination, repair and replication. The activity of RP-A is mediated by single-stranded DNA binding and protein interactions. Required for the efficient recruitment of the DNA double-strand break repair factor RAD51 to chromatin in response to DNA damage. Heterotrimer of 70, 32 and 14 kDa chains (canonical replication protein A complex). Component of the alternative replication protein A complex (aRPA) composed of RPA1, RPA3 and RPA4. The DNA-binding activity may reside exclusively on the 70 kDa subunit. Binds to SERTAD3/RBT1. Interacts with TIPIN. Directly interacts with PPP4R2, but not with SMEK2; this interaction is DNA damage-dependent and leads RPA2 dephosphorylation by PPP4C recruitment. Interacts with RAD51, preferentially when hyperphosphorylated. Directly interacts with RFWD3. 3 isoforms of the human protein are produced by alternative splicing.
