Heterotrimeric G proteins function to relay information from cell surface receptors to various intracellular effectors. G proteins comprise alpha, beta and gamma subunits, and following activation the alpha subunit binds GTP and dissociates from the betagamma complex. A large group of proteins have been identified as GTPase-activating proteins (GAPs), including the RGS (regulator of G protein signaling) family, which serve to deactivate specific Galpha isoforms by increasing the rate at which they convert GTP to GDP. A subfamily of RGS proteins expressed in the central nervous system contain, in addition to the highly conserved RGS domain, a characteristic GGL domain, or G protein gamma subunit-like domain, which mediates binding to Gbeta 5 subunits. This subfamily, which includes RGS6, RGS7, RGS9 and RGS11, associates with Gbeta 5 to form active GAP complexes that are predominantly localized to the cytosol. RGS/beta5 complexes preferentially target Galphao subunit for hydrolysis and inhibit Gbeta1gamma2-mediated activation of phospholipase C.

Uniprot Description

RGS7: inhibits signal transduction by increasing the GTPase activity of G protein alpha subunits thereby driving them into their inactive GDP-bound form. Activity on G(o)-alpha is specifically enhanced by the RGS6/GNG5 dimer. May play a role in synaptic vesicle exocytosis. May play important role in the rapid regulation of neuronal excitability and the cellular responses to short-lived stimulations. Heterodimer with GNG5. Interacts with RGS7BP, leading to regulate the subcellular location of the heterodimer formed with Gbeta5 (By similarity). Interacts with 14-3-3 protein Tau and SNAP25BP. Four alternatively spliced isoforms have been described.
