Tested: Human
Expected from sequence similarity: Human, Mouse, Rat, Pig
Western Blot: In transfected HEK293 transiently expressing Human PKD1 (DYKDDDDK tagged), a band of approx. 100kDa is observed. The same band is observed using anti- DYKDDDDK antibody and no bands are observed in cells transiently expressing Human PKD2 or PKD3. Recommended concentration 0.1-0.3ug/ml
Peptide ELISA: Antibody detection limit dilution 1:128000.
Western Blot (WB)
(HEK293 lysate overexpressing Human DYKDDDDK-tagged PKD1, Human DYKDDDDK-tagged PKD2 or Human GST-tagged PKD3 probed with (0.1ug/ml) in top panel, probed with)
Western Blot (WB)
(HEK293 lysate overexpressing Human DYKDDDDK-tagged PKD1 was used to immunoprecipitate PKD1 with 2ug The precipitate was subsequently probed in Western blot using at 1ug/ml. The secondary anti-goat picks up the heavy chain of used for the immunoprecipitation (annotated as IgG). Data kindly obtained from Dr Peter Storz, Mayo Clinic, USA)
NCBI and Uniprot Product Information
NCBI Description
PRKD1 is a serine/threonine kinase that regulates a variety of cellular functions, including membrane receptor signaling, transport at the Golgi, protection from oxidative stress at the mitochondria, gene transcription, and regulation of cell shape, motility, and adhesion (summary by Eiseler et al., 2009 [PubMed 19329994]).[supplied by OMIM, Nov 2010]
Uniprot Description
PRKD1: a CAMK kinase of the PKD family. Cleavage by caspase-3 following DNA damage activates it and alters its subcellular localization. Sensitizes cells to apoptosis induced by genotoxic stress. Its cleavage is blocked in cells that over-express the anti-apoptotic Bcl-x(L) protein. Expression of a caspase-resistant mutant partially inhibits DNA damage-induced apoptosis. Its activation by TLR ligands is dependent on MyD88, IRAK4 and -1, but not TRAF6. Essential for MyD88-dependent proinflammatory immune responses. Activated by diacylglycerol and phorbol esters. Binds to the trans-Golgi network and regulates the fission of transport carriers specifically destined to the cell surface. Colocalizes with F-actin at peripheral F-actin-rich structures in membrane ruffles at the edge of lamellipodia in cervical carcinoma cells. Substrates reportedly include critical regulatory proteins including CREB, SSH1L, CTNNB1, HDACs 5 and 7, PKD1, HPK1, MARK2, PIP5K2A, PPP1R14A

Product Notes
The PRKD1 prkd1 (Catalog #AAA423679) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The Goat Anti-PRKD1 (aa233-246) Antibody reacts with Human, Mouse, Rat, Pig Tested: Human Expected from sequence similarity: Human, Mouse, Rat, Pig and may cross-react with other species as described in the data sheet. AAA Biotech's PRKD1 can be used in a range of immunoassay formats including, but not limited to, Peptide ELISA (EIA), WB-Trf, Immunoprecipitation (IP). Immunoprecipitation: PKD1 was immunoprecipitated from Hek293T cells (as above). Immunoprecipitates were analyzed by Western Blot probed with the same antibody. Recommended concentration 0.1-0.3ug/ml Peptide ELISA: Antibody detection limit dilution 1:128000. Researchers should empirically determine the suitability of the PRKD1 prkd1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: QKSPSESFIG REKR. It is sometimes possible for the material contained within the vial of "PRKD1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
