NCBI Description

PhoPQ has been primarily characterized in Salmonella, with significant differences from Escherichia coli. [More information is available at EcoGene: EG10732]. The subunit structure in vivo is not known. [More information is available at EcoCyc: EG10732].

Uniprot Description

Function: Member of the two-component regulatory system PhoQ/PhoP involved in adaptation to low Mg2+ environments and the control of acid resistance genes. In presence of low periplasmic Mg2+ concentrations, PhoQ functions as a membrane-associated protein kinase that undergoes autophosphorylation and subsequently transfers the phosphate to PhoP, which results in the expression of PhoP-activated genes (PAG) and repression of PhoP-repressed genes (PRG). In presence of high periplasmic Mg2+ concentrations, acts as a protein phosphatase that dephosphorylates phospho-PhoP, which results in the repression of PhoP-activated genes and may lead to expression of some PhoP-repressed genes

By similarity. Mediates magnesium influx to the cytosol by activation of mgtA. Promotes expression of the two-component regulatory system rstA/rstB and transcription of the hemL, mgrB, nagA, slyB, vboR and yrbL genes. Ref.7

Catalytic activity: ATP + protein L-histidine = ADP + protein N-phospho-L-histidine.

Enzyme regulation: Acetyl-CoA acts as a non-competitive inhibitor of the PhoQ autokinase activity. Ref.9

Induction: PhoQ/phoP operon is positively autoregulated by both PhoP and PhoQ in a Mg2+-dependent manner. Ref.7 Ref.9 Ref.10 Ref.14

Miscellaneous: There is a close linkage between the PhoQ/PhoP and Rcs signaling systems, and both signaling systems respond to certain external divalent cations (zinc and magnesium).Two-component regulatory system EvgA/EvgS interacts with PhoQ/PhoP via signal transduction mediated by phospho-EvgA.
