Immunofluorescence (IF)

(Immunofluorescent analysis of PEA15 staining in HEK293T cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).)

NCBI Description

This gene encodes a death effector domain-containing protein that functions as a negative regulator of apoptosis. The encoded protein is an endogenous substrate for protein kinase C. This protein is also overexpressed in type 2 diabetes mellitus, where it may contribute to insulin resistance in glucose uptake. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2014]

Uniprot Description

PEA-15: an ubiquitously expressed anti-apoptotic protein. Inhibits both Fas- and Tnfrsf1a/Tnfr1-mediated caspase-8 activity and apoptosis. Interacts with CASP8/FLICE and FADD. Regulates glucose transport by controlling both the content of GLUT1 glucose transporters on the plasma membrane and the insulin-dependent trafficking of GLUT4 from the cell interior to the surface. Most abundant in tissues such as heart, brain, muscle and adipose tissue which utilize glucose as an energy source. Lower expression in glucose-producing tissues. Higher levels of expression are found in tissues from individuals with type 2 diabetes than in controls.

Protein type: Apoptosis; Motility/polarity/chemotaxis

Chromosomal Location of Human Ortholog: 1q21.1

Cellular Component: cytosol

Molecular Function: protein binding

Biological Process: activation of MAPK activity; MAPKKK cascade; negative regulation of glucose import; transport

Product Notes

The PEA15 pea15 (Catalog #AAA8248068) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-PEA15 Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's PEA15 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunofluorescence (IF), Immunocytochemistry (ICC). WB: 1/500 - 1/2000; Immunofluorescence/Immunocytochemistry: 1/50 - 1/100. Researchers should empirically determine the suitability of the PEA15 pea15 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PEA15, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
