Ordering
For Research Use Only!
Host
Rabbit
Reactivity
Mouse (Expected from Sequence), Rat (Expected from Sequence), Xenopus (Expected from Sequence)
Clonality
Polyclonal
Isotype
IgG
Specificity
This item detects a specific epitope within the carboxy-terminal of programmed cell death 4 (Pdcd4), a novel tumour suppressor. Pdcd4 interacts with the RNA helicase eIF4A and the scaffolding protein eIF4G, two components of the translation initiation complex, and thus inhibits protein synthesis. Pdcd4 has also been shown to inhibit transcription factor activator protein-1 (AP-1), which regulates several events required for cell invasion, including expression of matrix metalloproteinases and cell motility.

Pdcd4 localizes to the nucleus in proliferating cells, and its expression is modulated by cytokines in natural killer and T cells. Pdcd4 expression is down regulated with tumour progression in carcinomas of the lung, colon, prostate and breast.

Antiserum Preparation
Antisera to Pdcd4 (CT) were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared by affinity chromatography.
Target Species
Human
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 18 months from date of despatch.

NCBI Description

This gene is a tumor suppressor and encodes a protein that binds to the eukaryotic translation initiation factor 4A1 and inhibits its function by preventing RNA binding. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2010]

Uniprot Description

PDCD4: protein localized to the nucleus in proliferating cells that seems to possess a tumor suppressor activity. Directly interacts with the RNA helicase eIF4A and inhibits protein synthesis by interfering with the assembly of the cap-dependent translation initiation complex. Suppresses carbonic anhydrase type II protein expression in carcinoid cell lines. Since tumor cells require a high bicarbonate flux for their growth, carbonic anhydrase suppression results in growth inhibition. Expression of this gene is modulated by cytokines in natural killer and T cells. The gene product is thought to play a role in apoptosis but the specific role has not yet been determined. Two differentially spliced isoforms have been identified.

Protein type: Tumor suppressor; Apoptosis

Chromosomal Location of Human Ortholog: 10q24

Cellular Component: nucleoplasm; cytoplasm; cytosol; nucleus

Molecular Function: protein binding; RNA binding

Biological Process: negative regulation of JNK activity; apoptosis; cell aging; negative regulation of transcription, DNA-dependent; negative regulation of cell cycle

Product Notes

The PDCD4 pdcd4 (Catalog #AAA222849) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI HUMAN PDCD4 (C-TERMINAL) reacts with Mouse (Expected from Sequence), Rat (Expected from Sequence), Xenopus (Expected from Sequence) and may cross-react with other species as described in the data sheet. AAA Biotech's PDCD4 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Immunohistology Paraffin*, Western Blot (WB). Western Blot: This item detects a band of approximately 52kDa. ELISA: Minimum Dilution: 1/10000; Maximum Dilution: 1/50000 Immunohistology - Paraffin: Minimum Dilution: 1/100; Maximum Dilution: 1/300; Application Note: This product requires protein digestion pre-treatment of paraffin sections e.g. trypsin or pronase. Western Blotting: Minimum Dilution: 1/1000; Maximum Dilution: 1/10000. Researchers should empirically determine the suitability of the PDCD4 pdcd4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PDCD4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
