Immunofluorescence (IF) (Immunofluorescence staining of PBX1 in MCF7 cells. Cells were fixed with 4% PFA,blocked with 10% serum, and incubated with rabbit anti-human PBX1 polyclonal antibody (1:1000) at 4 degree C overnight. Then cells were stained with the Alexa Fluor594-conjugated Goat Anti-rabbit IgG secondary antibody (red). Positive staining was localized to nucleus.)

Related Product Information for anti-PBX1 antibody
PBX1 was abnormally overexpressed and its intracellular localization was found to be frequently amplified in many types of cancer, including renal clear cell carcinoma. PBX1 displays oncogenic activity in several different types of cells. PBX1 is a TALE homeodomain protein involved in the development of different types of cancers. Indeed, PBX1 is required for hematopoiesis as well as for multiple developmental processes such as skeletal patterning and organogenesis. PBX1 is a biological candidate gene for type 2 diabetes at the 1q21-q24 susceptibility locus.

NCBI Description

This gene encodes a nuclear protein that belongs to the PBX homeobox family of transcriptional factors. Studies in mice suggest that this gene may be involved in the regulation of osteogenesis, and required for skeletal patterning and programming. A chromosomal translocation, t(1;19) involving this gene and TCF3/E2A gene, is associated with pre-B-cell acute lymphoblastic leukemia. The resulting fusion protein, in which the DNA binding domain of E2A is replaced by the DNA binding domain of this protein, transforms cells by constitutively activating transcription of genes regulated by the PBX protein family. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Mar 2011]

Uniprot Description

PBX1: Binds the sequence 5'-ATCAATCAA-3'. Acts as a transcriptional activator of PF4 in complex with MEIS1. Converted into a potent transcriptional activator by the (1;19) translocation. May have a role in steroidogenesis and, subsequently, sexual development and differentiation. Forms a heterodimer with MEIS1 which binds DNA including a cAMP-responsive sequence in CYP17. Also forms heterotrimers with MEIS1 and a number of HOX proteins including HOXA9, HOXD4, HOXD9 and HOXD10. Interacts with PBXIP1. Expressed in all tissues except in cells of the B and T lineage. Belongs to the TALE/PBX homeobox family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The PBX1 pbx1 (Catalog #AAA8100507) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-PBX1 Antibody, Rabbit Polyclonal reacts with Human Predicted to React with: Mouse (Species predicted to react based on 100% sequence homology) and may cross-react with other species as described in the data sheet. AAA Biotech's PBX1 can be used in a range of immunoassay formats including, but not limited to, Immunocytochemistry (ICC), Immunofluorescence (IF). ICC/IF: 1:300-1:10,000. Researchers should empirically determine the suitability of the PBX1 pbx1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PBX1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
