Western Blot (WB) (Figure 1. Western blot analysis of PARP9 using anti-PARP9 antibody (MBS1753791).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: rat skeletal muscle tissue lysatesLane 2: mouse skeletal muscle tissue lysates.After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1. 5 hour at RT. The membrane was incubated with rabbit anti-PARP9 antigen affinity purified polyclonal antibody (Catalog # MBS1753791) at 0. 5 μg/mL overnight at 4 degree C, then washed with TBS-0. 1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1. 5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # MBS176460) with Tanon 5200 system. A specific band was detected for PARP9 approximately 130KD. The expected band size for PARP9 is at 96KD. )

Preparation and Storage
Store at -20 degree C for one year. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for a longer time. Avoid repeated freezing and thawing.

Poly(ADP-ribosyl)ation is a post-translational modification of proteins mediated by one of the 17 members of the poly(ADP-ribose) polymerases (PARP). PARP-9 belongs to the subfamily of macroPARPs, associating one to three macro domains to the PARP domain. Overexpression of PARP-9 stimulates cell migration in vitro, suggesting a role for PARP-9 in the promotion of malignant B cell migration and dissemination in high risk DLBCL. PARP-9 is also likely a transcription coactivator, its overexpression in B lymphocytes, stimulated by IFNgamma, inducing the transcription of IFNgamma-controlled genes.
