Western Blot (WB), Immunohistochemistry (IHC), Immunocytochemistry (ICC)
Application Notes
Suitable for use in Western Blot, Immunohistochemistry and Immunocytochemistry.
Dilution: Western Blot (Reducing): 1:1000. Cell membrane extracts were examined by SDS-PAGE under reducing conditions and transferred to a nylon membrane. Membranes were blocked for 1 hour at 4 C with PBS, 0.1% Tween 20, 2.5% milk powder. Membranes were incubated with diluted P1003-08B overnight at 4 C then rinsed with PBS and incubated with IgG (HRP) Goat anti-rat for 1 hour at RT. Following rinsing, the membranes were processed using ECL
Immunohistochemistry: 1:1000. Antiserum was used on perfusion fixed tissue. Perfusion: 1) calcium-free Tyrode's solution. 2) Paraformaldehyde-picric acid fixative. 3) PBS, 10% sucrose as a cryo-protectant. Desired tissues were dissected and stored overnight in PBS, 10% sucrose. Slide-mounted tissue sections were processed for Indirect Immunofluorescence. Slides were incubated with blocking buffer for 1 hour at RT. P1003-08B was diluted with blocking buffer to the appropriate working concentration. Blocking buffer was removed. Slides were incubated for 18-24 hours at 4 degree C with primary antiserum. Slides were rinsed 3 times with PBS. Slides were incubated with secondary antibodies for 1 hour at RT. Slides were again rinsed 3 times with PBS and coverslipped. Staining was examined using Fluorescence Microscopy.
Immunocytochemistry: 1:1000. P2X3 transfected cells were processed for Indirect Immunofluorescence. Media was removed. Cells were gently washed 3 times with serum-free media. Following fixation procedure, cells were processed for Indirect Immunofluorescence as described for Immunohistochemistry.
Immunogen
Synthetic peptide corresponding to aa383-397 VEKQSTDSGAYSIGH of rat P2X3.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

NCBI Description

The product of this gene belongs to the family of purinoceptors for ATP. This receptor functions as a ligand-gated ion channel and may transduce ATP-evoked nociceptor activation. Mouse studies suggest that this receptor is important for peripheral pain responses, and also participates in pathways controlling urinary bladder volume reflexes. It is possible that the development of selective antagonists for this receptor may have a therapeutic potential in pain relief and in the treatment of disorders of urine storage. Product Notes

The P2RX3 p2rx3 (Catalog #AAA613826) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The P2X3 Receptor reacts with Human, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's P2X3 Receptor can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC), Immunocytochemistry (ICC). Researchers should empirically determine the suitability of the P2RX3 p2rx3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "P2X3 Receptor, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
