NCBI Description

The OPRM1 gene encodes the mu opioid receptor, which is the primary site of action for the most commonly used opioids, including morphine, heroin, fentanyl, and methadone. It is also the primary receptor for endogenous opioid peptides beta-endorphin (see POMC, MIM 176830) and the enkephalins (see PENK, MIM 131330). The OPRM1 receptor is a membrane of the G protein-coupled receptor family (Bond et al., 1998 [PubMed 9689128]). There are at least 3 types of opioid receptors, mu, kappa (OPRK1; MIM 165196), and delta (OPRD1; MIM 165195), each with a distinct pharmacologic profile (Chen et al., 1993 [PubMed 8393525]).[supplied by OMIM]

Uniprot Description

Function: Inhibits neurotransmitter release by reducing calcium ion currents and increasing potassium ion conductance. Receptor for beta-endorphin.

Subunit structure: Forms a complex with G(alpha)z/i2 subunits and the RGSZ proteins, RGSZ17 and RGSZ20. The formation of this complex results in mu-opioid receptor desensitization. Interacts with RGSZ17 and RGSZ20

By similarity. The C-terminal tail interacts with the C-terminal section of DNAJB4. Ref.17

Subcellular location: Cell membrane; Multi-pass membrane protein.

Polymorphism: Variant Asp-40 does not show altered binding affinities for most opioid peptides and alkaloids tested, but it binds beta-endorphin, an endogenous opioid that activates the mu opioid receptor, approximately 3 times more tightly than the most common allelic form.

Sequence similarities: Belongs to the G-protein coupled receptor 1 family.
