NCBI Description

This gene encodes the second largest, catalytic subunit of the nuclear DNA-dependent RNA polymerase IV (aka RNA polymerase D). The NRPD2 protein is found at nuclear foci that overlap or are adjacent to chromocentromeres but are not fully coincident with chromocentromeres. The loss of NRPD2 leads to the loss of cytosine methylation at pericentromeric 5S genes and AtSN1 retroelements but has no discernible effect on centromere repeat methylation. This suggests that Pol IV primarily affects facultative heterochromatin rather than constitutive heterochromatin.

Uniprot Description

DNA-dependent RNA polymerase catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates. Second largest component of RNA polymerases IV and V which mediate short-interfering RNAs (siRNA) accumulation and subsequent RNA-directed DNA methylation-dependent (RdDM) transcriptional gene silencing (TGS) of endogenous repeated sequences, including transposable elements. Proposed to contribute to the polymerase catalytic activity and forms the polymerase active center together with the largest subunit. Also required for full erasure of methylation when the RNA trigger is withdrawn. Required for intercellular RNA interference (RNAi) leading to systemic post-transcriptional gene silencing. Involved in the maintenance of post-transcriptional RNA silencing. During interphase, mediates siRNA-independent heterochromatin association and methylation into chromocenters and condensation and cytosine methylation at pericentromeric major repeats. Required for complete maintenance of the 35S promoter homology-dependent TGS in transgenic plants and for the initial establishment of DNA methylation.

Product Notes

The NRPD2 nrpd2 (Catalog #AAA9009311) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Rabbit anti-Arabidopsis thaliana (Mouse-ear cress) NRPD2 Polyclonal Antibody reacts with Arabidopsis thaliana (Mouse-ear cress) and may cross-react with other species as described in the data sheet. AAA Biotech's NRPD2 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), WB (Western Blot) (ensure identification of antigen). Researchers should empirically determine the suitability of the NRPD2 nrpd2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "NRPD2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
