LXRa and LXRb (NR1H3 and NR1H2, respectively) are ligand-activated nuclear receptors that mediate cholesterol homeostasis. Once activated, LXRs upregulate the transcription of genes involved in cholesterol transport such as those encoding ATP-binding cassette (ABC) transporters, including ATP-binding cassette protein A (ABCA)1 and apolipoprotein E (Apo E), in various brain diseases. ABCA1 is known to carry and deliver cholesterol through receptor-mediated endocytosis, while in Alzheimer's brain, both ABCA1 and Apo E are shown to influence amyloid-beta transport and clearance. It is also shown that LXR ligands inhibit STAT1 binding to related gene promoters and the downstream expression of IRF1, TNFa, and IL-6. One study reports that differential SUMOlyation of LXRa and LXRb positively transrepresses STAT1 inflammatory signaling in IFN-g-stimulated rat brain astrocytes and that transient tranfection of specific individual siRNAs against LXRb and LXRa blocks LXR-induced suppression of IRF1 and TNFa expression.

NCBI Description

The liver X receptors, LXRA (NR1H3; MIM 602423) and LXRB, form a subfamily of the nuclear receptor superfamily and are key regulators of macrophage function, controlling transcriptional programs involved in lipid homeostasis and inflammation. The inducible LXRA is highly expressed in liver, adrenal gland, intestine, adipose tissue, macrophages, lung, and kidney, whereas LXRB is ubiquitously expressed. Ligand-activated LXRs form obligate heterodimers with retinoid X receptors (RXRs; see MIM 180245) and regulate expression of target genes containing LXR response elements (summary by Korf et al., 2009 [PubMed 19436111]).[supplied by OMIM, Jan 2010]
