Western Blot (WB)
(Figure 1. Western blot analysis of NOX5 using anti-NOX5 antibody. Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: human Hela whole cell lysate,Lane 2: human placenta tissue lysates,Lane 3: human SGC-7901 whole cell lysate,Lane 4: human Jurkat whole cell lysate,Lane 5: human THP-1 whole cell lysate. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-NOX5 antigen affinity purified polyclonal antibody at 0.5 ug/ml overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for NOX5 at approximately 86KD. The expected band size for NOX5 is at 86KD.)
Background: NOX5 (Nadph Oxidase 5), also known as NOX5A or NOX5B, is a protein which in humans is encoded by the NOX5 gene. The International Radiation Hybrid Mapping Consortium mapped the NOX5 gene to chromosome 15. NOX5 is a novel NADPH oxidase that generates superoxide and functions as an H+ channel in a Ca2+-dependent manner. It is found that, when heterologously expressed, NOX5 was quiescent in unstimulated cells. However, in response to elevations of the cytosolic Ca2+ concentration, it generated large amounts of superoxide. Using RT-PCR and Southern and Western blot analyses, NOX5 was identified as a flavin-containing Ca2+-dependent oxidase present in hairy leukemic cells (HC), but not normal B cells.
NCBI and Uniprot Product Information
NCBI Description
This gene is predominantly expressed in the testis and lymphocyte-rich areas of spleen and lymph nodes. It encodes a calcium-dependen NADPH oxidase that generates superoxide, and functions as a calcium-dependent proton channel that may regulate redox-dependent processes in lymphocytes and spermatozoa. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Oct 2011]
Uniprot Description
Calcium-dependent NADPH oxidase that generates superoxide. Also functions as a calcium-dependent proton channel and may regulate redox-dependent processes in lymphocytes and spermatozoa. May play a role in cell growth and apoptosis. Isoform v2 and isoform v5 are involved in endothelial generation of reactive oxygen species (ROS), proliferation and angiogenesis and contribute to endothelial response to thrombin.

Product Notes
The NOX5 nox5 (Catalog #AAA1751446) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-NOX5 Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's NOX5 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Direct ELISA (EIA). WB: 0.1-0.5 mug/ml Direct ELISA: 0.1-0.5 mug/ml Tested Species: In-house tested species with positive results. Researchers should empirically determine the suitability of the NOX5 nox5 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "NOX5, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
