Western Blot (WB)
(Western blot analysis of extracts from Myeloma cells, using Nociceptin Antibody. The lane on the left was treated with blocking peptide.Observed bands: 25 kDa.)
Western Blot (WB)
(Western blot analysis of extracts from Rat brain, using Nociceptin Antibody. The lane on the left was treated with blocking peptide.Observed bands: 25 kDa.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612144 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612144 1:200) and mouse anti-beta tubulin Ab(T0023 1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Post Translational Modifications: Specific enzymatic cleavages at paired basic residues probably yield other active peptides besides nociceptin. The N-terminal domain contains 6 conserved cysteines thought to be involved in disulfide bonding and/or processing.
Subcellular Location: Secreted.
Tissue Specificity: Predominantly expressed in the brain and spinal cord. Also expressed and secreted by peripheral blood neutrophils following degranulation.
Similarity: Belongs to the opioid neuropeptide precursor family.
NCBI and Uniprot Product Information
Predicted Molecular Weight: (Calculated)20kDa.
NCBI Description
This gene encodes a preproprotein that is proteolytically processed to generate multiple protein products. These products include nociceptin, nocistatin, and orphanin FQ2 (OFQ2). Nociceptin, also known as orphanin FQ, is a 17-amino acid neuropeptide that binds to the nociceptin receptor to induce increased pain sensitivity, and may additionally regulate body temperature, learning and memory, and hunger. Another product of the encoded preproprotein, nocistatin, may inhibit the effects of nociceptin. [provided by RefSeq, Jul 2015]
Uniprot Description
PNOC: Nociceptin is the ligand of the opioid receptor-like receptor (OPRL1). It may act as a transmitter in the brain by modulating nociceptive and locomotor behavior. May be involved in neuronal differentiation and development. Belongs to the opioid neuropeptide precursor family.

Product Notes
The PNOC pnoc (Catalog #AAA9612144) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Nociceptin Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig(100%), Bovine(100%), Horse(100%), Sheep(100%), Rabbit(100%), Dog(100%), Chicken(100%), Xenopus(100%) and may cross-react with other species as described in the data sheet. AAA Biotech's Nociceptin can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). WB: 1:500-1:2000 IF: 1:100-1:500 ICC: 1:100-1:500 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the PNOC pnoc for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Nociceptin, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
