Application Notes
IP: 4ul immunoprecipitated NF KB, p52 and its 110kDa precursor from 500ug of a Raji RIPA cell lysate.
WB: 1:1000 dilution detected NF KB, p52 and its 110kDa precursor in RIPA lysates from Raji cells. Raji cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-NF KB, p52 (1:1000). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
Applications are based on unconjugated antibody.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50), aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

NFkB is formed through the association of multiple subunits, either as a homodimer or heterodimer. Subunits have been identified as p50 (NFkB1), p65 (RelA), c-Rel, RelB and p52 (NFkB2). The classic NFkB form exists as a p50-p65 heterodimer and predominates in many cell types. Many of the possible combinatorial forms of homo-and heterodimers have been identified and growing evidence indicates that different forms of NFkB have different functions in cells. Interestingly, both the p50 and p52 subunits are derived from the precursor proteins p105 and p100 respectively, that each contain multiple copies of the so-called ankyrin repeat at their C-termini. Nuclear translocation of NFkB is confirmed by the use of electrophorectic mobility shift assays or by immunoblotting with nuclear extracts. The subunit composition of NFkB is confirmed by the use of antibodies that "supershift" the DNA/protein complex.

NCBI Description

This gene encodes one of the subunits of the transcription factor complex nuclear factor-kappa-B (NFkB). The NFkB transcription factor complex is expressed in numerous cell types and functions as a central activator of genes involved in inflammation and immune function. The NFkB complex can consist of different subunits that form both homo- or heterodimers which bind specific kappa-B elements in target genes. This gene encodes the p100 subunit that is processed into the active p52 subunit. This protein can function as both a transcriptional activator and repressor, depending on its dimer partner. Alternate splicing results in both coding and non-coding variants. [provided by RefSeq, May 2012]

Uniprot Description

NFkB-p100: a transcription factor of the nuclear factor-kappaB ( NFkB) group. Precursor of the p52 subunit of the nuclear factor NF-kappa-B, which binds to the kappa-B consensus sequence 5'-GGRNNYYCC-3', located in the enhancer region of genes involved in immune response and acute phase reactions. The precursor protein itself does not bind to DNA. LT-beta receptor agonists and LPS induce NF-kappaB/p100 processing to p52 at the level of the ribosome. There are five NFkB proteins in mammals (RelA/NFkB-p65, RelB, c-Rel, NF-_B1/NFkB-p105, and NF-_B2/NFkB-p100). They form a variety of homodimers and heterodimers, each of which activates its own characteristic set of genes. Two spliced isoforms have been identified. Isoform p49 is a subunit of the NF-kappa-B protein complex, which stimulates the HIV enhancer in synergy with p65.
