This immunoaffinity purified antibody recognizes a 50kD protein, corresponding to the apparent molecular masses of NF£eB, p50 on SDS-PAGE immunoblots, in samples from mouse and rat. Additional high molecular mass bands may sometimes be detected. These may respresent the p105 precursor protein. The antibody specificity is confirmed in Western blot by peptide competition studies.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50), aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Nuclear factor-kappa B, also known as NFkappaB, KBF1, p110 and EBP-1, was identified as a sequence specific transcriptional activator that binds to the intronic enhancer of kappa light chain gene in B lymphocytes (1,2). NF-kappaB is a heterodimer that consists of a 50kD DNA binding subunit (p50) and a 65 kD transactivation subunit (p65/Rel A) (3). Both of these subunits exhibit sequence homology to the protooncogene c-Rel (4). The p50 has an isoform called p49/p52, and both proteins are derived from the amino-terminal of precursor protein p105 (5,6) and p100 (7), respectively. The p50/p65 heterodimer remains in the cytosol in an inactive form as a complex with its inhibitor, IkappaB. Upon stimulation of cells by a wide variety of stimuli such as lipopolysaccharide (LPS), pro-inflammatory cytokines (interleukin-1, tumor necrosis factor), and viral infection, IkappaB is phosphorylated and degraded by proteosome. The active NF-kappaB heterodimer is translocated into the nucleus and will induce gene expression.

NCBI Description

This gene encodes a 105 kD protein which can undergo cotranslational processing by the 26S proteasome to produce a 50 kD protein. The 105 kD protein is a Rel protein-specific transcription inhibitor and the 50 kD protein is a DNA binding subunit of the NF-kappa-B (NFKB) protein complex. NFKB is a transcription regulator that is activated by various intra- and extra-cellular stimuli such as cytokines, oxidant-free radicals, ultraviolet irradiation, and bacterial or viral products. Activated NFKB translocates into the nucleus and stimulates the expression of genes involved in a wide variety of biological functions. Inappropriate activation of NFKB has been associated with a number of inflammatory diseases while persistent inhibition of NFKB leads to inappropriate immune cell development or delayed cell growth.
