Immunogen
A synthetic peptide corresponding to a sequence in the middle region of human SIRT2(165-187aa YTQNIDTLERIAGLEQEDLVEAH), both different from the related mouse and rat sequences by three amino acids.
Preparation and Storage
At -20 degree C for one year. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for a longer time. Avoid repeated freezing and thawing.

Background: SIRT2 is a predominantly cytoplasmic protein that colocalizes with microtubules. This gene encodes a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. SIRT2 is mapped to 19q13.2. It is able to cause radioactivity to be transferred from (32P) NAD to bovine serum albumin (BSA). According to researches, SIRT2 can act as a bona fide tubulin deacetylase. SIRT2 was found to deacetylate lys40 of alpha-tubulin both in vitro and in vivo. SIRT2 is also a regulator of mitotic progression that acts downstream from CDC14B in a pathway regulating mitotic exit or subsequent cytokinesis. What's more, SIRT2 is an important regulator of programmed necrosis.

NCBI Description

This gene encodes a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have not yet been determined; however, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. Studies suggest that the human sirtuins may function as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity. The protein encoded by this gene is included in class I of the sirtuin family. Several transcript variants are resulted from alternative splicing of this gene. [provided by RefSeq, Jul 2010]

Involved in the control of mitotic exit in the cell cycle, probably via its role in the regulation of cytoskeleton. Deacetylates PCK1, opposing proteasomal degradation. Deacetylates 'Lys-310' of RELA. Interacts with HDAC6, suggesting that these proteins belong to a large complex that deacetylate the cytoskeleton. Widely expressed. Highly expressed in heart, brain and skeletal muscle, while it is weakly expressed in placenta and lung. Down-regulated in many gliomas suggesting that it may act as a tumor suppressor gene in human gliomas possibly through the regulation of microtubule network. Inhibited by Sirtinol, A3 and M15 small molecules. Inhibited by nicotinamide. Belongs to the sirtuin family. Class I subfamily. 4 isoforms of the human protein are produced by alternative splicing.
