The MYO7A gene encodes a protein belonging to the unconventional myosin superfamily. Mutations in the MYO7A gene are associated with Usher Syndrome type 1B (USH1B) and nonsyndromic hearing loss (DFNB2, DFNA11). Mutations within MYO7A can lead to either non-syndromic hearing loss or the Usher syndrome type 1B (USH1B). Most mutations in the MYO7A gene caused USH1B, whereas only a few reported mutations led to DFNB2 and DFNA11. MYO7A is expressed in human photoreceptors and retinal pigment epithelium. In the retina, most MYO7A is localized in the apical region of the RPE (retinal pigmented epithelial) cells, where many of the reactions of the visual retinoid cycle take place, and a small amount is associated with the ciliary and periciliary membranes of the photoreceptor cells. Its roles appear to be quite varied.
