Principle of the Assay: As mentioned above, this kit utilizes the Double Antibody Sandwich ELISA technique. The pre-coated antibody is an anti-Rat MBP monoclonal antibody, while the detection antibody is a biotinylated polyclonal antibody. Samples and biotinylated antibodies are added into ELISA plate wells and washed out with PBS or TBS after their respective additions to the wells. Then Avidin-peroxidase conjugates are added to the wells in after. TMB substrate is used for coloration after the enzyme conjugate has already been thoroughly washed out of the wells by PBS or TBS. TMB reacts to form a blue product from the peroxidase activity, and finally turns to yellow after addition of the stop solution (Color Reagent C). The color intensity and quantity of target analyte in the sample are positively correlated.

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with MBP. Standards and samples are pipetted into the wells with a biotin-conjugated MBP and avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MBP antibody bound in the initial step. The color development is stopped and the intensity of the color is measured.

Principle of the Assay: MBP ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MBP antibody and an MBP-HRP conjugate. The assay sample and buffer are incubated together with MBP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MBP concentration since MBP from samples and MBP-HRP conjugate compete for the anti-MBP antibody binding site. Since the number of sites is limited, as more sites are occupied by MBP from the sample, fewer sites are left to bind MBP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MBP concentration in each sample is interpolated from this standard curve.<br><br>Intended Uses: This MBP ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse MBP. Principle of the Assay: MBP ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for MBP. Standards or samples are then added to the microtiter plate wells and MBP if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of MBP present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for MBP are added to each well to "sandwich" the MBP immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain MBP and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MBP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MBP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MBP is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MBP bound in the initial step. The color development is stopped and the intensity of the color is measured.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Capture antibody was pre-coated onto 96-well plates. And the biotin conjugated antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the target amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of target can be calculated.

Intended Uses: This immunoassay kit allows for the in vitro quantitative determination of target antigen concentrations in serum, plasma, tissue homogenates, cell culture supernates or other biological fluids.<br><br>Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to target antigen. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for target antigen and then avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain target antigen, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm.
