Specificity
This item recognises an epitope within the central region (IN) of human Myeloid cell leukaemia sequence 1 (MCL-1), an anti-apoptotic member of the Bcl-2 family of proteins, first identified in differentiating ML-1 myeloid cells, and expressed in many different cell lines, tissues and a variety of tumours.

The MCL-1 protein, localised to intracellular membranes and particularly mitochondrial membranes, has since been isolated in two distinct isoforms, known as MCL-1 long (MCL-1L) and MCL-1 short (MCL-1S). MCL-1S has been identified as a pro-apoptotic splice variant of MCL-1L, missing the transmembrane and BH1 and BH2 (Bcl-2 homology) domains. Interaction between these two antagonistic variants can act to regulate cell survival.

This item recognises both the MCL-1L (38kDa) and MCL-1S (30kDa) isoforms of MCL-1.

NCBI Description

This gene encodes an anti-apoptotic protein, which is a member of the Bcl-2 family. Alternative splicing results in multiple transcript variants. The longest gene product (isoform 1) enhances cell survival by inhibiting apoptosis while the alternatively spliced shorter gene products (isoform 2 and isoform 3) promote apoptosis and are death-inducing. [provided by RefSeq, Oct 2010]

Uniprot Description

MCL1: a myeloid cell leukemia protein of the Bcl-2 family of proteins. Two alternatively spliced transcripts encoding distinct isoforms have been identified. The longer gene product (isoform 1) enhances cell survival by inhibiting apoptosis while the alternatively spliced shorter gene product (isoform 2) promotes apoptosis and is death-inducing.

Product Notes

The Mcl-1 mcl1 (Catalog #AAA221875) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's Mcl-1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Western Blot: This item detects a band of approximately 30kDa and 38kDa in Human Raji cell lysates. Western Blotting: Minimum Dilution: 0.5; Maximum Dilution: 1.0ug/ml. Researchers should empirically determine the suitability of the Mcl-1 mcl1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Mcl-1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
