NCBI Description

The product of this gene functions in the NF-kappaB signal transduction pathway. The encoded protein, and the similar and functionally redundant protein MAP3K7IP2/TAB2, forms a ternary complex with the protein kinase MAP3K7/TAK1 and either TRAF2 or TRAF6 in response to stimulation with the pro-inflammatory cytokines TNF or IL-1. Subsequent MAP3K7/TAK1 kinase activity triggers a signaling cascade leading to activation of the NF-kappaB transcription factor. The human genome contains a related pseudogene. Alternatively spliced transcript variants have been described, but their biological validity has not been determined. [provided by RefSeq, Jul 2008]

Uniprot Description

TAB3: Adapter linking MAP3K7/TAK1 and TRAF6 or TRAF2. Mediator of MAP3K7 activation, respectively in the IL1 and TNF signaling pathways. Plays a role in activation of NF-kappa-B and AP1 transcription factor. Isoform 2 may be an oncogenic factor. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Adaptor/scaffold

Chromosomal Location of Human Ortholog: Xp21.2

Cellular Component: cytoplasm; cytosol; endosome membrane; nucleoplasm; plasma membrane

Molecular Function: protein binding; zinc ion binding

Biological Process: activation of MAPK activity; activation of NF-kappaB transcription factor; I-kappaB kinase/NF-kappaB cascade; JNK cascade; MyD88-dependent toll-like receptor signaling pathway; negative regulation of autophagy; stimulatory C-type lectin receptor signaling pathway

Product Notes

The MAP3K7IP3 tab3 (Catalog #AAA420409) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The Goat anti-MAP3K7IP3 (aa 59 to 69) Antibody reacts with Expected from sequence similarity: Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's MAP3K7IP3 can be used in a range of immunoassay formats including, but not limited to, Peptide ELISA (EIA). Peptide ELISA: Antibody detection limit dilution 1: 32000. Western Blot: Preliminary experiments gave an approx 100kDa band in Human Kidney lysates after 0.5ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 78.7kDa according to NP_690000.2. The 100kDa band was successfully blocked by incubation with the immunizing peptide. Researchers should empirically determine the suitability of the MAP3K7IP3 tab3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: HSPDDNRMNR N. It is sometimes possible for the material contained within the vial of "MAP3K7IP3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
