Application Notes
Suitable for use in ELISA and Western Blot.
Dilution: Direct ELISA: 0.15-0.3ug/ml using 100ul/well antibody solution. Allows the detection of at least 0.2ng/well of recombinant rat MIP-1a.
Western Blot: 0.1-0.2ug/ml. The dectection limit is 1.5-3ng/lane, under reducing or non-reducing conditions.

Highly purified (>98%) recombinant rat Macrophage Inflammatory Protein 1 alpha (MIP-1 alpha).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Both MIP-1 alpha and MIP-1 beta are structurally and functionally related CC chemokines. They participate in the host response to invading bacterial, viral, parasite and fungal pathogens by regulating the trafficking and activation state of selected subgroups of inflammatory cells e.g. macrophages, lymphocytes and NK cells. While both MIP-1 alpha and MIP-1 beta exert similar effects on monocytes their effect on lymphocytes differ; with MIP-1 alpha selectively attracting CD8+ lymphocytes and MIP-1 beta selectively attracting CD4+ lymphocytes. Additionally, MIP-1 alpha and MIP-1 beta have also been shown to be potent chemoattractants for B cells, eosinophils and dendritic cells. Both human and murine MIP-1 alpha and MIP-1 beta are active on human and murine hematopoietic cells.

NCBI Description

This locus represents a small inducible cytokine. The encoded protein, also known as macrophage inflammatory protein 1 alpha, plays a role in inflammatory responses through binding to the receptors CCR1, CCR4 and CCR5. Polymorphisms at this locus may be associated with both resistance and susceptibility to infection by human immunodeficiency virus type 1.

Recombinant MIP-1-alpha induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). Belongs to the intercrine beta (chemokine CC) family.
