EDG4 belongs to a family of G-protein coupled receptors whose ligands are lysophospholipids. The ligand for EDG4 is lysophospholipid. There are 6 known members of the EDG receptor family and they are implicated in mediating growthrelated effects such as induction of cellular proliferation, alterations in differentiation and survival and suppression of apoptosis. They also evoke cellular effector functions that are dependent on cytoskeletal responses such as contraction, secretion, adhesion and chemotaxis. EDG receptors are developmentally regulated and differ in tissue distribution. They couple to multiple types of G proteins to signal through ras and MAP kinase, rho, phospholipase C and several protein tyrosine kinases. EDG4 is expressed in testes, ovarian tumor and leukocyte containing tissues.

NCBI Description

This gene encodes a member of family I of the G protein-coupled receptors, as well as the EDG family of proteins. This protein functions as a lysophosphatidic acid (LPA) receptor and contributes to Ca2+ mobilization, a critical cellular response to LPA in cells, through association with Gi and Gq proteins. An alternative splice variant has been described but its full length sequence has not been determined. [provided by RefSeq, Jul 2008]

Function: Receptor for lysophosphatidic acid (LPA), a mediator of diverse cellular activities. Seems to be coupled to the G(i)/G(o), G12/G13, and G(q) families of heteromeric G proteins. Plays a key role in phospholipase C-beta (PLC-beta) signaling pathway. Stimulates phospholipase C (PLC) activity in a manner that is independent of RALA activation. Ref.9 Ref.11

Subunit structure: Interacts with SLC9A3R2/NHERF2, MAGI3 and PLCB3. Interacts with RALA and ADRBK1. Ref.9 Ref.10 Ref.11

Subcellular location: Cell surface. Cell membrane; Multi-pass membrane protein. Note: Prior to LPA treatment found predominantly at the cell surface but in the presence of LPA co-localizes with RALA in the endocytic vesicles. Ref.11

Tissue specificity: Expressed most abundantly in testes and peripheral blood leukocytes with less expression in pancreas, spleen, thymus and prostate. Little or no expression in heart, brain, placenta, lung, liver, skeletal muscle, kidney, ovary, small intestine, or colon.

Miscellaneous: Ref.1 cDNA clone has a guanine nucleotide deletion that causes a frameshift near its C-terminal coding region. This likely reflects a somatic mutation in the ovary tumor cells from which the cDNA was isolated and may have altered the function of the encoded receptor, and contributed to transformation of the original ovary cells that formed a tumor.
