Alexa Fluor 488-conjugated goat anti-rabbit lgG at 1:400 dilution was used as the secondary antibody (green). Cytoplasmic actin was counterstained with Dylight Fluor 554 (red) conjugated Phalloidin (red).)
Western Blot (WB)
(LIN28A Antibody (Center) western blot analysis in mouse Neuro-2a cell line lysates (35ug/lane).This demonstrates the LIN28A antibody detected the LIN28A protein (arrow).)
Immunofluorescence (IF)
(Fluorescent confocal image of SY5Y cells stained with MBS9201994 LIN28A (Center) antibody. SY5Y cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min), then incubated with MBS9201994 LIN28A (Center) primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (5.25 uM, 25 min). Lin28a immunoreactivity is localized very specifically to the nuclei of the SY5Y cells.)
Immunohistochemistry (IHC)
(LIN28A Antibody (Center) immunohistochemistry analysis in formalin fixed and paraffin embedded human testis tissue followed by peroxidase conjugation of the secondary antibody and DAB staining. This data demonstrates the use of LIN28A Antibody (Center) for immunohistochemistry. Clinical relevance has not been evaluated.)
Flow Cytometry (FC/FACS)
(LIN28A Antibody (Center) flow cytometric analysis of Hela cells (right histogram) compared to a negative control cell (left histogram).FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.)

NCBI and Uniprot Product Information
NCBI Description
This gene encodes a LIN-28 family RNA-binding protein that acts as a posttranscriptional regulator of genes involved in developmental timing and self-renewal in embryonic stem cells. The encoded protein functions through direct interaction with target mRNAs and by disrupting the maturation of certain miRNAs involved in embryonic development. This protein prevents the terminal processing of the LET7 family of microRNAs which are major regulators of cellular growth and differentiation. Aberrant expression of this gene is associated with cancer progression in multiple tissues. [provided by RefSeq, Sep 2015]
Uniprot Description
LIN28A: Acts as a 'translational enhancer', driving specific mRNAs to polysomes and thus increasing the efficiency of protein synthesis. Its association with the translational machinery and target mRNAs results in an increased number of initiation events per molecule of mRNA and, indirectly, in stabilizing the mRNAs. Binds IGF2 mRNA, MYOD1 mRNA, ARBP/36B4 ribosomal protein mRNA and its own mRNA. Essential for skeletal muscle differentiation program through the translational up-regulation of IGF2 expression. Acts as a suppressor of microRNA (miRNA) biogenesis by specifically binding the precursor let-7 (pre-let- 7), a miRNA precursor. Acts by binding pre-let-7 and recruiting ZCCHC11/TUT4 uridylyltransferase, leading to the terminal uridylation of pre-let-7. Uridylated pre-let-7 miRNAs fail to be processed by Dicer and undergo degradation. Degradation of pre- let-7 in embryonic stem (ES) cells contributes to the maintenance of ES cells. In contrast, LIN28A down-regulation in neural stem cells by miR-125, allows the processing of pre-let-7. Specifically recognizes the 5'-GGAG-3' motif in the terminal loop of pre-let-7. Also recognizes and binds non pre-let-7 pre-miRNAs that contain the 5'-GGAG-3' motif in the terminal loop, leading to their terminal uridylation and subsequent degradation. Monomer. During skeletal muscle differentiation, associated with translation initiation complexes in the polysomal compartment. Directly interacts with EIF3S2. Interaction with NCL is RNA-dependent. Interacts with ZCCHC11/TUT4. Can be negatively regulated by the interaction of microRNAs miR-125a and miR-125b with at least two miRNA responsive elements (miREs) in the 3'-UTR of this gene. These interactions may reduce both translation efficiency and mRNA abundance. Negatively regulated by retinoic acid. Expressed in embryonic stem cells (ES cells), placenta and testis. Belongs to the lin-28 family.

The immunogen sequence is 108-138. The LIN28A Antibody (Center) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's LIN28A can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), ELISA (EIA), Immunofluorescence (IF), Immunohistochemistry (IHC), Flow Cytometry (FC/FACS). IF~~1:10~50 WB~~1:1000. Researchers should empirically determine the suitability of the LIN28A lin28a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "LIN28A, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
