This product recognises human leptin, a 16KDa major adipose-derived hormone and member of the leptin family, which plays a key role in appetite control and the regulation of energy expenditure.Secreted leptin associates with proteins within the bloodstream and is transported to the brain, where it inhibits or up-regulates the activity of neuropeptides such as neuropeptide Y (NPY) and alpha-melanocyte-stimulating hormone respectively. Leptin is a recognised marker of obesity, with a correlation between circulating levels of leptin and body fat mass, and defects in leptin production result in severe obesity. Regulators of leptin expression include insulin, cortisol, cAMP and thiazolidinediones.

Application Notes
ELISA: This purified human leptin antibody may be used in an indirect ELISA or as the capture reagent in a sandwich ELISA with a biotinylated human leptin antibody as the detection reagent and recombinant human leptin as the standard.
Western Blot: This purified human leptin antibody may be used in Western Blotting applications under either reducing or non-reducing conditions with recombinant human leptin as the positive control.
ELISA: Minimum Dilution: 0.5; Maximum Dilution: 2.0ug/ml
Immunohistology - Paraffin: Maximum Dilution: 0.5ug/ml; Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose.
Western Blotting: Minimum Dilution: 0.1; Maximum Dilution: 0.2ug/ml
Reconstitution
Reconstitute with 0.1ml distilled water Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution. For long term storage the addition of 0.09% sodium azide is recommended.

Antiserum to human leptin was raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.

Preparation and Storage
Prior to reconstitution store at 4 degree C. After reconstitution store at -20 degree C. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of reconstitution.

NCBI Description

This gene encodes a protein that is secreted by white adipocytes, and which plays a major role in the regulation of body weight. This protein, which acts through the leptin receptor, functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing. Mutations in this gene and/or its regulatory regions cause severe obesity, and morbid obesity with hypogonadism. This gene has also been linked to type 2 diabetes mellitus development. Defects in LEP may be a cause of obesity (OBESITY). It is a condition characterized by an increase of body weight beyond the limitation of skeletal and physical requirements, as the result of excessive accumulation of body fat.
