Testing Data

(Published customer image: Rabbit anti Human laminin antibody used for the visualization of laminin in normal colon and in colonic adenocarcinoma tissue sections by immunofluorescence. Image caption:Representative indirect immunofluorescence images showing double-stained sections for the detection of the alpha1 integrin subunit and laminin in a normal colon mucosa (A and B) and a moderately differentiated colon adenocarcinoma specimen (C, D). The alpha1 subunit (red signal) was observed at the basolateral domain of normal epithelial cells of the lower crypt compartment (LC in panel A and E in panel B) compared to weak signal in the upper compartment (UC, panel A). In CRC, alpha1 was also localized at the basolateral domain of tumour epithelial cells (E) as well as in the adjacent subepithelial myofibroblasts (MF) (Panels C, D). Basement membrane (arrows) located at the interface between the two tissues was stained with an anti-laminin antibody (green staining). Nuclei were stained with DAPI (blue). Scale bar?=?50 um.From: Boudjadi S, Carrier JC, Beaulieu JF.Integrin alpha1 subunit is up-regulated in colorectal cancer.Biomark Res. 2013 Mar 7;1(1):16.)

NCBI Description

Laminins, a family of extracellular matrix glycoproteins, are the major noncollagenous constituent of basement membranes. They have been implicated in a wide variety of biological processes including cell adhesion, differentiation, migration, signaling, neurite outgrowth and metastasis. Laminins are composed of 3 non identical chains: laminin alpha, beta and gamma (formerly A, B1, and B2, respectively) and they form a cruciform structure consisting of 3 short arms, each formed by a different chain, and a long arm composed of all 3 chains. Each laminin chain is a multidomain protein encoded by a distinct gene. Several isoforms of each chain have been described. Different alpha, beta and gamma chain isomers combine to give rise to different heterotrimeric laminin isoforms which are designated by Arabic numerals in the order of their discovery, i.e. alpha1beta1gamma1 heterotrimer is laminin 1. The biological functions of the different chains and trimer molecules are largely unknown, but some of the chains have been shown to differ with respect to their tissue distribution, presumably reflecting diverse functions in vivo. This gene encodes the beta chain isoform laminin, beta 1. The beta 1 chain has 7 structurally distinct domains which it shares with other beta chain isomers. The C-terminal helical region containing domains I and II are separated by domain alpha, domains III and V contain several EGF-like repeats, and domains IV and VI have a globular conformation. Laminin, beta 1 is expressed in most tissues that produce basement membranes, and is one of the 3 chains constituting laminin 1, the first laminin isolated from Engelbreth-Holm-Swarm (EHS) tumor. A sequence in the beta 1 chain that is involved in cell attachment, chemotaxis, and binding to the laminin receptor was identified and shown to have the capacity to inhibit metastasis. Uniprot Description

Binding to cells via a high affinity receptor, laminin is thought to mediate the attachment, migration and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components. Involved in the organization of the laminar architecture of cerebral cortex. It is probably required for the integrity of the basement membrane/glia limitans that serves as an anchor point for the endfeet of radial glial cells and as a physical barrier to migrating neurons. Radial glial cells play a central role in cerebral cortical development, where they act both as the proliferative unit of the cerebral cortex and a scaffold for neurons migrating toward the pial surface.

Product Notes

The LAMB1 lamb1 (Catalog #AAA225133) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Rabbit anti Laminin reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Laminin can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). WB: 1:1000-1:4000. Researchers should empirically determine the suitability of the LAMB1 lamb1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Laminin, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
