Related Product Information for anti-KPNA6 antibody
KPNA6 Antibody: Karyopherin, a cytosolic and heterodimeric protein complex consisting of alpha and beta subunits, is responsible for targeting proteins with nuclear localization signals to the nuclear pore complex (NPC) by an energy requiring, Ran-dependent mechanism. The alpha subunit and imported substrate enter the nucleus and accumulate in the nucleoplasm, while the beta subunit accumulates at the NPC. KPNA6 belongs to a subfamily within the KPNA family that also includes IPOA5&6. Down-regulation of KPNA6 by RNAi expression in HeLa cells strongly inhibited cell proliferation, possibly due to blocking the nuclear import of specific factors essential for cell growth and proliferation. KPNA6 also interacts with the Rev protein of HIV-1 and promote its nuclear import.

NCBI Description

Nucleocytoplasmic transport, a signal- and energy-dependent process, takes place through nuclear pore complexes embedded in the nuclear envelope. The import of proteins containing a nuclear localization signal (NLS) requires the NLS import receptor, a heterodimer of importin alpha and beta subunits also known as karyopherins. Importin alpha binds the NLS-containing cargo in the cytoplasm and importin beta docks the complex at the cytoplasmic side of the nuclear pore complex. In the presence of nucleoside triphosphates and the small GTP binding protein Ran, the complex moves into the nuclear pore complex and the importin subunits dissociate. Importin alpha enters the nucleoplasm with its passenger protein and importin beta remains at the pore. The protein encoded by this gene is a member of the importin alpha family. [provided by RefSeq, Jul 2008]

Uniprot Description

KPNA6: Functions in nuclear protein import as an adapter protein for nuclear receptor KPNB1. Binds specifically and directly to substrates containing either a simple or bipartite NLS motif. Docking of the importin/substrate complex to the nuclear pore complex (NPC) is mediated by KPNB1 through binding to nucleoporin FxFG repeats and the complex is subsequently translocated through the pore by an energy requiring, Ran- dependent mechanism. At the nucleoplasmic side of the NPC, Ran binds to importin-beta and the three components separate and importin-alpha and -beta are re-exported from the nucleus to the cytoplasm where GTP hydrolysis releases Ran from importin. The directionality of nuclear import is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus. Belongs to the importin alpha family.

The KPNA6 Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's KPNA6 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Western Blot (WB), Immunofluorescence (IF). KPNA6 antibody can be used for detection of KPNA6 by Western blot at 1 ug/mL. Antibody can also be used for immunofluorescence starting at 20 ug/mL.For immunofluorescence start at 20 ug/mL.

Optiaml dilutions for each applciation to be determined by the researcher. Researchers should empirically determine the suitability of the KPNA6 kpna6 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "KPNA6, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
