Dilution: Western Blot: 0.1-0.2ug/ml. The detection limit is ~5ng/lane under non-reducing and reducing conditions.
Flow Cytometry: 50ug/ml, 10ul labels 1-2.5x10e5 MG-63 human osteosarcoma cells in a total reaction volume <200ul. The binding of unlabeled antibodies may be visualized by adding 10ul of a 25ug/ml stock solution of a secondary developing reagent such as anti-sheep lgG conjugated to a fluorochrome.
Immunocytochemistry: 10ug/ml detects Integrin, beta5 in MG-63 human osteosarcoma cells. Cells were fixed with PBS, 4% paraformaldehyde for 20 minutes at RT and blocked with PBS, 10% normal donkey serum, 0.1% Triton X-100, 1% BSA for 45 minutes at RT. After blocking, cells were incubated with diluted I7661-43E overnight at 4 degree C followed by Rhodamine Red coupled anti-sheep IgG at RT in the dark for one hour. Between each step, cells were washed with PBS, 0.1% BSA.
Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~0.2ng/well.
