Each vial contains 5mg BSA, 0.9mg NaCl, 0.2mg Na2HPO4, 0.05mg Thimerosal, 0.05mg NaN3.
Reconstitution
Add 0.2ml of distilled water will yield a concentration of 500ug/ml.
Preparation and Storage
At -20 degree C for one year. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for a longer time. Avoid repeated freezing and thawing.

Background: Nitric oxide synthase, inducible is an enzyme that in humans is encoded by the NOS2 gene. Nitric oxide(NO) is a messenger molecule with diverse functions throughout the body. In the brain and peripheral nervous system, NO displays many properties of a neurotransmitter; it is implicated in neurotoxicity associated with stroke and neurodegenerative diseases, neural regulation of smooth muscle, including peristalsis, and penile erection. Three different NOS isoforms have been identified which fall into two distinct types, constitutive and inducible. The inducible NOS(iNOS) isoform is expressed in a variety of cell types and tissues in response to inflammatory agents and cytokines. The human iNOS(NOS2) gene is approximately 37 kb in length and consists of 26 exons and 25 introns. Diefenbach et al.(1999) studied the relationship of IL12 and nitric oxide synthase-2(NOS2) to innate immunity to the parasite Leishmania in mice. And conclude that NOS2-derived NO is a prerequisite for cytokine signaling and function in innate immunity. From studies in Tanzania and Kenya, Hobbs et al.(2002) identified a novel single-nucleotide polymorphism, -1173C-T(163730.0001), in the NOS2 promoter that was significantly associated with protection from symptomatic malaria and severe malarial anemia.

Nitric oxide is a reactive free radical which acts as a biologic mediator in several processes, including neurotransmission and antimicrobial and antitumoral activities. This gene encodes a nitric oxide synthase which is expressed in liver and is inducible by a combination of lipopolysaccharide and certain cytokines. Three related pseudogenes are located within the Smith-Magenis syndrome region on chromosome 17. [provided by RefSeq, Jul 2008]

Also has nitrosylase activity and mediates cysteine S-nitrosylation of cytoplasmic target proteins such COX2. Homodimer. Binds SLC9A3R1. By endotoxins and cytokines. Induced by IFNG/IFN-gamma acting synergistically with bacterial lipopolysaccharides (LPS), TNF or IL1B/interleukin-1 beta. Expressed in the liver, retina, bone cells and airway epithelial cells of the lung. Not expressed in the platelets. Regulated by calcium/calmodulin. Aspirin inhibits expression and function of this enzyme and effects may be exerted at the level of translational/post-translational modification and directly on the catalytic activity. Belongs to the NOS family. 2 isoforms of the human protein are produced by alternative splicing.
