Western Blot (WB) (Figure 1. Western blot analysis of IL10 using anti-IL10 antibody. Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: human U-87MG whole cell lysate,Lane 2: rat testis tissue lysates,Lane 3: mouse testis tissue lysates,Lane 4: mouse SP20 whole cell lysate. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-IL10 antigen affinity purified polyclonal antibody at 0.5 ug/ml overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for IL10 at approximately 19KD. The expected band size for IL10 is at 21KD.)

Related Product Information for anti-IL10 antibody
Description: Rabbit IgG polyclonal antibody for IL10 detection. Tested with WB, Direct ELISA in Human; Mouse; Rat.
Background: Interleukin-10 (IL-10 or IL10), also known as human cytokine synthesis inhibitory factor (CSIF), is an anti-inflammatory cytokine. In humans IL-10 is encoded by the IL10 gene. It is capable of inhibiting synthesis of pro-inflammatory cytokines like IFN-gamma, IL-2, IL-3, TNFalpha and GM-CSF made by cells such as macrophages and regulatory T-cells.IL-10 also displays potent abilities to suppress the antigen presentation capacity of antigen presenting cells. Kim et al. (1992) showed that the mouse Il 10 gene contains 5 exons and spans about 5.2 kb of genomic DNA. Eskdale et al. (1997) mapped the IL10 gene to the junction between 1q31 and 1q32.

NCBI Description

The protein encoded by this gene is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract. Mutations in this gene are associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis.[provided by RefSeq, May 2011]

Uniprot Description

Inhibits the synthesis of a number of cytokines, including IFN-gamma, IL-2, IL-3, TNF and GM-CSF produced by activated macrophages and by helper T-cells.

Product Notes

The IL10 il10 (Catalog #AAA1751198) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-IL10 Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's IL10 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Direct ELISA (EIA). WB: 0.1-0.5 mug/ml Direct ELISA: 0.1-0.5 mug/ml Tested Species: In-house tested species with positive results. Researchers should empirically determine the suitability of the IL10 il10 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IL10, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
