Immunofluorescence (IF)/Immunocytochemistry (ICC)

(MBS9612191 staining A549 cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612191) and mouse anti-beta tubulin Ab(T0023) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody.The nuclear counter stain is DAPI (blue).)

Function: Regulator that plays a central role in regulation of apoptosis and transcription via its interaction with NF-kappa-B and p53/TP53 proteins. Blocks transcription of HIV-1 virus by inhibiting the action of both NF-kappa-B and SP1. Also inhibits p53/TP53 function, possibly by preventing the association between p53/TP53 and ASPP1 or ASPP2, and therefore suppressing the subsequent activation of apoptosis (PubMed:12524540).

Subcellular Location: Cytoplasm. Nucleus. Note: Predominantly cytoplasmic but also nuclear.

Tissue Specificity: Highly expressed in heart, placenta and prostate. Weakly expressed in brain, liver, skeletal muscle, testis and peripheral blood leukocyte.

Subunit Structure: Interacts with RELA NF-kappa-B subunit and with SP1 via its C-terminus part. Interacts (via SH3 domain and ANK repeats) with p53/TP53; the interaction inhibits pro-apoptotic activity of p53/TP53 (PubMed:12524540). Interacts with TP63 and TP73.

Similarity: The N-terminal region is required for cytoplasmic localization.The ANK repeats and the SH3 domain are required for specific interactions with p53/TP53.Belongs to the iASPP family.

IASPP is one of the most evolutionarily conserved inhibitors of p53 (TP53; MIM 191170), whereas ASPP1 (MIM 606455) and ASPP2 (MIM 602143) are activators of p53.[supplied by OMIM, Mar 2008]

PPP1R13L: an oncoprotein that interacts with and inhibits NF-kappaB, Sp1 and p53. Plays a central role in regulation of apoptosis and transcription. May inhibit p53 by preventing its association with ASPP1 or ASPP2, thereby suppressing the subsequent activation of apoptosis. Required for neuronal survival after axonal injury. Two alternatively spliced isoforms have been described. Isoform 1 is preferrentially found in the cytoplasm; isoform 2 is nuclear.
