Immunohistochemistry (IHC)
(Anti-HIF3A antibody IHC of human lung, respiratory epithelium. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody concentration 5 ug/ml.)
Immunohistochemistry (IHC)
(Anti-HIF3A antibody IHC of human placenta. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody concentration 5 ug/ml.)
Western Blot (WB)
(Anti-Hif Antibody - Western Blot. Western blot of Affinity Purified anti-Hif antibody shows detection of a band ~72 kD corresponding to mouse Hif3a (arrowhead). Approximately 10 ug of a CoCl2 treated 3T3 cell lysate (lane 1) and control 3T3 cell lysate (lane 2) were separated by 4-20% SDS-PAGE and transferred onto nitrocellulose. Treatment of exponentially growing 3T3 cells with 130 uM CoCl2 for 6 h at 37? C effectively mimics hypoxia. After blocking the membrane was probed overnight at 4C with the primary antibody diluted to 1:1600. The membrane was washed and reacted with a 1:10000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX (for 45 min at room temperature. IRDye800 fluorescence image was captured using the Odyssey Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.)
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is the alpha-3 subunit of one of several alpha/beta-subunit heterodimeric transcription factors that regulate many adaptive responses to low oxygen tension (hypoxia). The alpha-3 subunit lacks the transactivation domain found in factors containing either the alpha-1 or alpha-2 subunits. It is thought that factors containing the alpha-3 subunit are negative regulators of hypoxia-inducible gene expression. Multiple alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Mar 2011]
Uniprot Description
HIF3A: Involved in adaptive response to hypoxia. Suppresses hypoxia-inducible expression of HIF1A and EPAS1. Binds to core DNA sequence 5'-TACGTG-3' within the hypoxia response element (HRE) of target gene promoters. The complex HIF3A-ARNT activates the transcription of reporter genes driven by HRE. Isoform 4 has a dominant-negative function of inactivating HIF1A-mediated transcription. Isoform 4 attenuates the binding of HIF1A to hypoxia-responsive elements (HRE), thus inhibiting HRE-driven transcription. Hypoxia induces down-regulation of isoform 4, leading to activation of HIF1A in hypoxia. Conversely, upon restoring normoxia, the expression of isoform 4 increases and thereby secure an inhibition of HIF1A activity. Isoform 4 may be a negative regulator of hypoxia-inducible gene expression in the kidney and may be involved in renal tumorigenesis. Functions as an inhibitor of angiogenesis in the cornea. 7 isoforms of the human protein are produced by alternative splicing.

The Rabbit Polyclonal to Mouse HIF3A / HIF3-Alpha reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's HIF3A / HIF3-Alpha can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC - Paraffin), Immunofluorescence (IF), Western Blot (WB), ELISA (EIA). ELISA (1:14000 - 1:80000), IF, IHC-P (5 ug/ml), WB (1:100 - 1:8000) Usage: Immunohistochemistry: was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antib.. Researchers should empirically determine the suitability of the HIF3A hif3a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "HIF3A / HIF3-Alpha, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
