Preparation and Storage
Store product at 4 degree C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20 degree C. Aliquots are stable at -20 degree C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Huntington's disease (HD), a neurodegenerative disorder characterized by loss of striatal neurons, is caused by an expansion of a polyglutamine tract in the HD protein huntingtin. HAP1 was initially identified through a two-hybrid library screening; the binding of HAP1 to huntingtin correlated with the expansion of the polyglutamine tract. HAP1 also interacts with two cytoskeletal proteins (dynactin and pericentriolar autoantigen protein 1), suggesting that HAP1 may play a role in vesicular trafficking or organelle transport. HAP1 is also involved with the huntingtin-enhanced BDNF transport along the cellular microtubles. Attenuation of this process led to the loss of neurotrophic support and neuronal toxicity, which suggests that loss of this function might contribute to pathogenesis. Several alternatively spliced isoforms have been described for HAP1.

NCBI Description

Huntington's disease (HD), a neurodegenerative disorder characterized by loss of striatal neurons, is caused by an expansion of a polyglutamine tract in the HD protein huntingtin. This gene encodes a protein that interacts with huntingtin, with two cytoskeletal proteins (dynactin and pericentriolar autoantigen protein 1), and with a hepatocyte growth factor-regulated tyrosine kinase substrate. The interactions with cytoskeletal proteins and a kinase substrate suggest a role for this protein in vesicular trafficking or organelle transport. Several alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Jul 2008]
