Application Notes
Suitable for use in ELISA, Western Blot and Immunohistochemistry.
Dilution: ELISA: 0.5-1.0ug/ml. The detection limit for rhHAND2 and rmHAND2 is approximately 1.3ng/well.
Western Blot: 0.1-0.2ug/ml. The detection limit for rhHAND2 and rmHAND2 is approximately 25ng/lane under non-reducing and reducing conditions.
Immunohistochemistry: 10ug/ml to detect HAND2 in mouse embryos. Cells were fixed with PBS containing 4% paraformaldehyde and blocked with PBS containing 10% normal donkey serum, 0.1% Triton X-100, and 1% BSA. After blocking, cells were incubated with diluted primary antibody followed by NorthernLightsTM 557-coupled anti-goat IgG at RT in the dark. Between each step, cells were washed with PBS containing BSA.

E. coli-derived rhHAND2.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute by adding sterile 40-50% glycerol, PBS. Aliquot and store at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

HAND2 (also known as dHAND, Thing-2 and Hed) is a 24kD, class B member of the basic helix-loop-helix transcription factors superfamily. HAND2 is essential for cardiac morphogenesis and required for vascular development and regulation of angiogenesis, possibly through VEGF signaling. HAND2 is also implicated in limb development acting as an upstream regulator of sonic hedgehog. Alternative initiation and internal deletions result in at least three potential HAND2 isoforms. Human and mouse HAND2 proteins are 100% identical.

NCBI Description

The protein encoded by this gene belongs to the basic helix-loop-helix family of transcription factors. This gene product is one of two closely related family members, the HAND proteins, which are asymmetrically expressed in the developing ventricular chambers and play an essential role in cardiac morphogenesis. Working in a complementary fashion, they function in the formation of the right ventricle and aortic arch arteries, implicating them as mediators of congenital heart disease. In addition, this transcription factor plays an important role in limb and branchial arch development. [provided by RefSeq]

HAND2: Essential for cardiac morphogenesis, particularly for the formation of the right ventricle and of the aortic arch arteries. Required for vascular development and regulation of angiogenesis, possibly through a VEGF signaling pathway. Plays also an important role in limb development, particularly in the establishment of anterior-posterior polarization, acting as an upstream regulator of sonic hedgehog (SHH) induction in the limb bud. Is involved in the development of branchial arches, which give rise to unique structures in the head and neck. Binds DNA on E-box consensus sequence 5'-CANNTG-3'.
