Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

GRB7 family adaptor proteins consist of GRB7, GRB10 and GRB14, which contain an N-terminal proline-rich SH3 binding domain, followed by PH, PBS and SH2 domains. Each member of the family has several splicing isoforms (1). It has been reported that GRB10 interacts with many receptor tyrosine kinases (RTKs) as well as downstream signal molecules such as Raf, Akt and Nedd4 (1,2). Although it was originally reported that GRB10 is exclusively phosphorylated at serine residues (3), recent research has shown that GRB10 is also phosphorylated at tyrosine 67 by Src kinase family members (4).

NCBI Description

The product of this gene belongs to a small family of adapter proteins that are known to interact with a number of receptor tyrosine kinases and signaling molecules. This gene encodes a growth factor receptor-binding protein that interacts with insulin receptors and insulin-like growth-factor receptors. Overexpression of some isoforms of the encoded protein inhibits tyrosine kinase activity and results in growth suppression. This gene is imprinted in a highly isoform- and tissue-specific manner, with expression observed from the paternal allele in the brain, and from the maternal allele in the placental trophoblasts. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq]

GRB10: an adaptor protein that inhibits insulin-stimulated IRS-PI (3)K-Akt signaling pathway by disrupting the association of IRS with the insulin receptor. Also binds activated PDGFR and EGFR. Preferentially localizes to the mitochondria, modulating the anti-apoptotic activity of mitochondrial Raf-1. Can translocate to the plasma membrane and lamellipodia. Three alternatively spliced isoforms have been described.
