Application Notes
IP: 4ug immunoprecipitated Flt3 from 500ug of mouse Bafl-3 RIPA lysate.
Applications are based on unconjugated antibody.
ICC: IgG, H&L (FITC) Pab Gt xRb
WB: 0.5-2ug/ml detected Flt3 in RIPA lysates from HeLa cells and mouse Bafl-3 cells. Bafl-3 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with (1ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.

Preparation and Storage
Store product at 4 degree C in the dark. DO NOT FREEZE! Stable at 4 degree C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.

Function: Tyrosine-protein kinase that acts as cell-surface receptor for the cytokine FLT3LG and regulates differentiation, proliferation and survival of hematopoietic progenitor cells and of dendritic cells. Promotes phosphorylation of SHC1 and AKT1, and activation of the downstream effector MTOR. Promotes activation of RAS signaling and phosphorylation of downstream kinases, including MAPK1/ERK2 and/or MAPK3/ERK1. Promotes phosphorylation of FES, FER, PTPN6/SHP, PTPN11/SHP-2, PLCG1, and STAT5A and/or STAT5B. Activation of wild-type FLT3 causes only marginal activation of STAT5A or STAT5B.
